Aydın, Balıkesir ve İzmir illerinde zeytin ağaçlarındaki viral hastalık etmenlerinin tanılanması ve bulunma durumlarının belirlenmesi

Öz

Bu çalışma 2009 ve 2010 yılları arasında Aydın,
Balıkesir ve İzmir illerinde yaygın olarak zeytin yetiştiriciliği yapılan
üretim alanlarındaki virüs hastalıklarının belirlenmesi ve toplanan örneklerde
virüslerin bulunma durumlarının ortaya konması amacıyla 2009 yılında Aydın
(157) ve İzmir (136) illerinden 293 örnek, 2010 yılında ise Balıkesir ilinden
82 örnek olmak üzere toplam 375 örnek alınarak yürütülmüştür. Zeytin
örneklerindeki viral etmenlerin tanılanması biyolojik, serolojik ve moleküler
yöntemler kullanılarak yapılmıştır. Arabis mosaic virus (ArMV), Cucumber
mosaic virus (CMV), Cherry leafroll virus (CLRV) ve Strawberry
latent ringspot virus (SLRSV) adlı etmenlere ait tanı kitleri kullanılarak,
örnekler DAS-ELISA yöntemine göre testlenmiştir. Bu virüslere ek olarak,
örneklerde Olive latent-1 virus (OLV-1), Olive latent-2 virus
(OLV-2),Olive latent-3 virus (OLV-3), Olive latent ringspot virus
(OLRSV) ve Olive leaf yellowing-associated virus (OLYaV) adlı virüslerin
varlığı RT-PCR yöntemi ile araştırılmıştır. RT-PCR sonuçları, 2009 ve 2010
yıllarında Aydın, Balıkesir ve İzmir illerinden
toplanarak
testlenen toplam 375 bitki örneğinde %55.73 oranında viral
enfeksiyon bulunduğunu ve bu oranın Aydın ilinde %22.66, Balıkesir ilinde %11.46
ve İzmir ilinde ise %21.60 düzeyinde olduğunu ortaya koymuştur. DAS-ELISA
sonuçlarına göre; örneklerin %12,80’inde ArMV ve sadece 1 örnekte ise SLRSV
enfeksiyonunun var olduğu ve test edilen diğer virüslerin ise bulunmadığı saptanmıştır.
RT-PCR testi sonucunda, örneklerin %22,93’ünde ArMV, %9,60’ında CMV,
%10,66’sında CLRV ve %9,06’sında SLRSV enfeksiyonlarının var olduğu görülmüştür.
Buna ek olarak, 9 örnekte CMV+ CLRV, 4 örnekte ise ArMV+CLRV karışık enfeksiyon
şeklinde tespit edilirken, tüm örneklerde OLV-1, OLV-2, OLV-3, OLRSV ve OLYaV’nin
bulunmadığı belirlenmiştir.

The identification of virus diseases in olive trees in Aydin, Balikesir and İzmir provinces and the determination of their present status

Öz

The present study was carried out to determine the presence and
prevalence of the viral agents in olive production areas of Aydın, Balıkesir and
Izmir in 2009-2010. In 2009, totally 293 samples were collected from Aydın
(157) and Izmir (136) provinces. In 2010, with collected 82 samples from
Balıkesir, the total sample number in the research has reached to 375. To
identify viral agents, the biological, serological and molecular methods were
applied to collected olive samples. DAS-ELISA method was used to detect Arabis
mosaic virus (ArMV), Cucumis mosaic virus (CMV), Cherry leafroll
virus (CLRV) and Strawberry latent ringspot virus (SLRSV). Also, the
existence of Olive latent-1 virus (OLV-1), Olive latent-2 virus
(OLV-2), Olive latent-3 virus (OLV-3), Olive latent ringspot virus
(OLRSV) and Olive leaf yellowing-associated virus (OLYaV) in the samples
was
analysed by RT-PCR
molecular method. The results of RT-PCR analyses showed that the incidence of
virus infection was 55.73%in total 375 olive samples from Aydın, Balıkesir and
Izmir in 2009 and 2010.
According to individual provinces, it was 22.66% in
Aydın, 11.46 in Balıkesir and 21.60% in Izmir.
DAS-ELISA results revealed that infection rate of ArMV was 12.8% in
total samples and only 1 sample was found to be infected with SLRSV while other
tested viruses were not present. According to the data from RT-PCR tests,
infection rates in all tested samples were 22.93%, 9.60%, 10.66% and 9.06% for
ArMV, CMV, CLRV and SLRSV, respectively. Moreover, it was ascertained that 9
samples were infected by CMV + CLRV and 4 samples by ArMV + CLRV as mixed
infections. Other viruses including OLV-1, OLV-2, OLV-3, OLRSV and OLYaV were
not detected in all samples.
