Cytotoxicity of fissure sealants in vitro

Öz

OBJECTIVE: This in vitro study aimed to evaluate the cytotoxicity of three different fissure sealant materials.

MATERIALS AND METHOD: Cylindrical-shaped (2x5 mm) fissure sealant test samples were prepared according to the manufacturers' directions (Fuji Triage, Dyract Seal, UltraSeal XT Plus). L929 mouse fibroblast cells were plated in 96-well plates, and maintained in an incubator at a mixture of 5% CO2/95% air and at 37 °C for 24 h. The test samples were immersed in Basal Medium Eagle (BME) with 10% New Born Calf Serum and 5% penicillin/streptomycin. After 24 h, the incubation medium of the cells was replaced by the medium in which the test samples were extracted. Cells that were not exposed to the material extracts were used as control. After 24 h, cell viability was determined by using MTT assay. The data were analyzed by One-Way ANOVA and Tukey's HSD Post-hoc tests.

RESULTS: Dyract Seal and Fuji Triage were significantly different from the control (p<0.05). UltraSeal XT Plus was similar to the control (p>0.05). Dyract Seal presented the greatest cytotoxic effect on the L929 cells.

CONCLUSION: The results of this study indicate that fissure sealant materials can release toxic agents. Biocompatibility, like for other restorative materials, is an important aspect for fissure sealants.

Fissür örtücülerin in vitro sitotoksisitesi

Öz

AMAÇ: Bu in vitro çalışmada üç farklı fissür örtücü materyalin sitotoksisitesinin değerlendirilmesi amaçlandı.

GEREÇ VE YÖNTEM: Üretici firmanın önerileri dikkate alınarak silindirik şekilli (2x5 mm) fissür örtücü test örnekleri hazırlandı (Fuji Triage, Dyract Seal, UltraSeal XT Plus). L929 fare fibroblast hücreleri 96-kuyucuklu hücre kültürü plakları içerisine yerleştirildi ve bir inkübatör içerisinde 24 sa. süre ile 37 °C’de, %5 CO2/%95 hava karışımlı bir ortamda bekletildi. Test örnekleri ise %10 New Born Calf Serum ve %5 penisilin/streptomisin içeren Basal Medium Eagle (BME) içerisine alındı. L929 hücrelerinin inkübasyon medyumu 24 sa. sonra uzaklaştırıldı ve yerine içerisinde 24 sa. boyunca test örneklerinin ekstrakte edildiği medyum konuldu. Test materyali içeren medyuma maruz bırakılmayan hücreler kontrol grubunu oluşturdu. Hücre canlılığı 24. sa. sonunda MTT testi ile belirlendi. Veriler One-Way ANOVA ve Tukey's HSD Post-hoc testleri ile analiz edildi.

BULGULAR: Dyract Seal ve Fuji Triage, kontrol gurubundan farklı idi (p<0.05). UltraSeal XT Plus ise kontrol grubu ile benzer hücre canlılığı gösterdi (p>0.05). Dyract Seal, L929 hücreleri üzerine en fazla sitotoksik etkiyi gösteren materyaldi.

SONUÇ: Bu çalışmanın sonuçları fissür örtücülerin bazı toksik ajanlar salabildiğini göstermektedir. Biyouyumluluk, diğer restoratif materyaller gibi fissür örtücüler için de kritik bir özelliktir.
