Experimental Toxoplasmosis in White Turkeys

Öz

In this
study, forming of experimental toxoplasmosis in white turkeys; clinical,
pathological and serological determination of tissue lesions, aimed to compare
them and determine pathogenesis. For these purposes, a total of 120 two months
old white turkeys were divided into groups as oral and parenteral infections
and also their controls. The oral group was infected with 0.5 mL of Toxoplasma
gondii RH strain inoculum suspension contains 10⁶ tachyzoites while
its control group was given 0.5 mL of saline. The parenteral group were divided
into four groups as intraperitoneal (IP), intramuscular (IM), intravenous (IV)
and cloacal (C). Each parenteral route was divided into two groups and one
control group for inoculums given 10⁵ and 10⁴ tachyzoites
in 0.5 mL. These two control groups were also given 0.5 mL saline as indicated
above. Due to acute toxoplasmosis, death occurred in three white turkeys given
tachyzoites IP 10⁵ showed neurological clinical symptoms as
torticollis, ataxia, and tremor. In the histopahologic examination of these
three turkeys, T. gondii tissue cysts were detected in the brain. Also, one of
given 10⁵ tachyzoites IP group in the brain and one given 10⁴
tachyzoites IV group in the liver were observed tissue cysts associated with
toxoplasmosis. The turkeys in all infection groups were found seropositive in
both Sabin-Feldman Dye Test (SFDT) and Indirect Hemagglutination Test (IHA).
The statistical difference between SFDT and IHA was insignificantly for the
both parenteral infection groups (P>0.05) while the difference was found
significantly for the orally infected group (P<0.05). In conclusion in the
present study, the tissue cysts of T. gondii were microscopically seen in brain
and liver of the experimental infected white turkeys.

Beyaz Hindilerde Deneysel Toksoplazmozis

Öz

Bu çalışmada, beyaz
hindilerde deneysel toxoplasmozis oluşturulması; klinik, patolojik ve serolojik
teknikler kullanılarak doku lezyonlarının belirlenmesi, bunların
karşılaştırılması ve patogenezi amaçlanmıştır. Çalışmada, 2 aylık yaşta 120
adet beyaz hindi oral ve parenteral yollarla ve aynı zamanda onların kontrol
grupları olarak gruplandırılmıştır. Oral gruba 0,5 mL’sinde 10⁶
takizoit içeren inokulum verilirken bunun kontrol grubuna 0,5 mL serum
fizyolojik verilmiştir. Parenteral grup; İntravenöz (İV), İntraperitoneal (İP),
İntramuskuler (İM), ve Kloakal (K) olarak dört gruba ayrılmıştır. Her bir parenteral
yol da 0,5 mL’sinde 10⁵ ve 10⁴ takizoit içeren inokulum
verilmesi için kendi içerisinde iki gruba ve birer kontrol grubuna ayrılmıştır.
Bunların kontrol gruplarınada 0,5 mL serum fizyolojik verilmiştir. İP 10⁵
takizoit verilen grubtaki üç hindide akut toxoplazmozise bağlı tortikollis,
ataksi ve tremor gibi sinirsel klinik belirtileri takiben ölüm şekillenmiştir.
Bu hindilerin histopatolojisinde beyinde doku kistleri izlenmiştir. Ayrıca İP
10⁵ takizoit verilen gruptaki bir hindide beyinde, İV 10⁴
takizoit verilen grubtaki bir hindide karaciğerde toksoplasmozis’e ilişkin doku
kisti görülmüştür. Sabin-Feldman Dye Test (SFDT) ve İndirekt Hemaglutinasyon (İHA)
ile 10⁴ ve 10⁵ takizoit gruplarında seropozitiflik
belirlenmiştir. SFDT ve İHA arasındaki istatistiksel fark önemsiz bulunmuştur
(P>0,05). 10⁶ takizoit verilerek enfekte edilen oral enfeksiyon
grubunda SFDT ve İHA arasında istatistiksel farklılık önemli bulunmuştur. (P<0,05).
Sonuç olarak bu çalışmada deneysel enfekte edilen beyaz hindilerde mikroskopik
olarak T. gondii doku kistleri beyin
ve karaciğerde görülmüştür.
