Keçi Sütlerinde Listeria spp. Prevalansı ve Virülent Listeria monocytogenes’in Real-Time PCR ile Belirlenmesi

Öz

Keçi sütü tüm dünyada yaygın olarak tüketilen koyun ve
inek sütüne alternatif bir gıdadır.           
Bu araştırmada Şanlıurfa ilinin Akçakale ve Eyyübiye ilçelerinde bulunan
küçük ölçekli işletmelerde üretilen çiğ keçi sütü örneklerinde virülent L. monocytogenes prevalansını belirlemek
amaçlanmıştır. 88 adet çiğ keçi sütü örneği altı farklı işletmeden
toplanmıştır. L. monocytogenes
suşları ISO 11290-1 metodu ile izole edilmiş ve hlyA gen bölgesine spesifik primer kullanılarak Real-time PCR
metodu ile identifiye edilmiştir. Araştırmada analiz edilen 88 adet çiğ keçi
sütü örneğinden 4’ü Listeria spp. (%4.54)
olarak izole edilmiş, bunlardan 2’si (%2.27) virulent L. monocytogenes olarak identifiye edilmiştir. Bu araştırma keçi
sütlerinin L. monocytogenes ile
kontamine olduğunu, ısıl işlem ve hijyen kurallarına dikkat edilmesi
gerektiğini vurgulamaktadır.

Prevalance of Listeria spp. and Detection of Virulent Listeria monocytogenes in Goat Milk by Real-time PCR

Öz

Goat milk is an alternative food to sheep and cow milk which is widely consumed all over the world.
In this study, it was aimed to determine the prevalence of virulent L. monocytogenes in raw goat milk
samples produced in small scale farms located in Akçakale and Eyyubiye districts of Şanlıurfa province.
88 samples of raw goats’ milk were collected from six different farms. L. monocytogenes strains were
isolated by the method of ISO 11290-1 and identified by Real-time PCR method using primer specific to
hlyA gene region. Of the 88 raw goat milk samples analyzed in the study, 4 (4.54 %) were Listeria spp.,
and 2 (2.27 %) of them were identified as virulent L. monocytogenes. This study emphasizes that goat’s
milk is contaminating with L. monocytogenes and heat treatment and hygiene rules should be considered.
