Antepfıstığı (Pistacia vera L.) Bahçelerinde Kök Çürüklüğü ve Solgunluğa Neden Olan Fungal Etmenlerin Araştırılması: Siirt İli Lokasyonu, Türkiye

Öz

Antepfıstığı (Pistacia vera L.) bahçelerinde son yıllarda kök çürüklüğü ve solgunluk gibi belirtiler gözlemlenmiş ve ağaçlarda kurumalar tespit edilmiştir. Bu çalışmada kurumalara neden olan toprak kökenli fungal patojenler araştırılmıştır. Çalışma; Siirt ili Merkez, Eruh, Şirvan, Kurtalan ilçeleri ve köylerinde 2021-2022 yıllarında güdümlü örnekleme sörvey metoduna göre yürütülmüştür. Elde edilen fungus türleri, renk, gelişim durumları, spor yapıları gibi morfolojik karakterlerine ve ITS (Internal Transcribed Spacer) gen dizilerinin NCBI (The National Center for Biotechnology Information) gen bankasındaki diğer genlerle eşleştirme analizleri ile belirlenmiştir. Patojenisite çalışması, bir yaşındaki Siirt çeşidi üzerinde yapılmış ve patojen izolatlar belirlenmiştir. Çalışma sonucunda ağırlıklı olarak Fusarium türleri (F. solani, F. oxysporum, F. verticillioides, F. equseti, F. acuminatum, F. avenaceaum, F. proliferatum, F. brchygibbosum, Fusarium spp.) ile Diaporthe hongkongensis, Scydalidium sp., Rosellinia sp., Nectria diminuta, Neoscytalidium dimitatum, Rhizoctonia solani, Macrophomina phaseolina, Phoma spp. ve Cylindrocarpon spp., izole edilmiş ve bunların farklı oranlarda patojen oldukları tespit edilmiştir.

Investigation of Fungal Causal Agents Root Rot and Wilt on Pistachio (Pistacia vera L.) Orchard: Siirt Province Location, Türkiye

Öz

In recent years, symptoms such as root rot and wilt have been observed in pistachio (Pistacia vera L.) orchards in this province, and drying of trees has been detected. This study investigated soil-borne fungal pathogens that cause diseases in crops. Survey studies were carried out according to the controlled sampling survey method in Siirt province center, Eruh, Şirvan, Kurtalan districts and villages in 2021-2022. Fungal species were determined based on morphological characters such as color, growth in nutrient media, and spore structures, and by matching analysis of Internal Transcribed Spacer (ITS) gene sequences with other genes in the National Center for Biotechnology Information (NCBI) gene bank. Pathogenicity study was carried out on one-year-old Siirt variety in the greenhouse and pathogenic isolates were determined. As a result of the study, mostly Fusarium species (F. solani, F. oxysporum, F. verticillioides, F. equseti, F. acuminatum, F. avenaceaum, F. proliferatum, F. brchygibbosum, Fusarium spp.) and Diaporthe hongkongensis, Scydalidium sp., Rosellinia sp., Nectria diminuta, Neoscytalidium dimitatum, Rhizoctonia solani, Macrophomina phaseolina, Phoma spp., and Cylindrocarpon spp. were isolated and some of them found to be pathogenic at different rates.
