Bu çalışma, erişkin kedi ovaryumlarında ilk defa TGF-α’nın immunohistokimyasal yerleşiminin belirlenmesi amacıyla yapıldı. Çalışmada 10 adet, evcil, erişkin kedi ovaryumu kullanıldı. Dokular %10’luk tamponlanmış nötr formol solüsyonunda tespit edildi ve rutin histolojik prosedür uygulandı. Kesitlere Crossman’ın üçlü boyaması uygulanarak ovaryumların genel histolojik yapısı ve indirekt streptavidin-biotin-peroksidaz kompleks yöntemi uygulanarak TGF-α’nın immunohistokimyasal yerleşimi incelendi. Ovaryumlarda TGF-α’ya ait pozitif boyamaların sadece granüloza hücrelerinde, teka interna hücrelerinde, folliküllerin ovositlerinde ve teka lutein hücrelerinde olduğu görüldü. Primordiyal folliküllerin ovositlerinde, büyük antral ve atretik foliküllerin granüloza hücrelerinde şiddetli immun reaksiyon gözlendi. Folliküllerin çevresini saran teka interna katmanında ve teka lutein hücrelerinde ise orta şiddetli immun reaksiyon gözlendi. Sonuç olarak, TGF-α’nın erişkin kedi ovaryumlarında parakrin ya da otokrin bir düzenleyici olarak hem folikülogeneziste hem de granüloza hücrelerinin proliferasyonunda ve differansiyasyonunda rol oynayacağı sonucuna varıldı. N2 - Transforming growth factor alpha (TGF-α) is a growth factor that plays important roles in the maturation of oocytes, proliferation, and differentiationof granulosa cells in ovaries. This study was performed to determine the immunohistochemical localization of TGF-α in adultcats’ ovaries for the first time. Ten domestic breed, adult cats’ ovaries were used in this study. Tissues were fixed in 10% neutral bufferedformalin solution and routine histological procedure was performed. Crossman's triple staining was applied to the sections to examine thehistologic structure of the ovaries and immunohistochemical localization of TGF-α was examined using the indirect streptavidin-biotin-peroxidasecomplex method. Positive staining for TGF-α in ovaries was observed only in granulosa cells, theca interna cells, oocytes, and thecalutein cells. Intense immunoreaction was observed in oocytes of primordial follicles and granulosa cells of large antral and atretic follicles. Amoderate immunoreaction was observed in the layer of theca interna surrounding the follicles and in theca lutein cells. In conclusion, it wasconcluded that TGF-α may play a role in both folliculogenesis and proliferation and, also differentiation of granulosa cells as a paracrine orautocrine regulator in adult cat ovaries.
