For this aim, canola plants showing severe mosaic symptoms were collected from 2 districts (Ezine and Lapseki) of Çanakkale province in Turkey. Collected twenty-one samples were tested by DAS-ELISA with polyclonal antisera (Bioreba, Switzerland) and RT-PCR using gene specific primers containing partial NIb+CP to determine the presence of Turnip mosaic virus (TuMV). DAS-ELISA and RT-PCR test results indicated that 15 sample were infected with TuMV. Corresponding 1178 bp DNA fragment of isolate CK01 chosen randomly was purified and sequenced. The nucleotide sequence of the partial NIb+CP gene of CK01 isolate was compared with TuMV isolates from different parts of the world found in gen bank. Sequence analysis of partial NIb+CP genes showed 88-93% identities among CK01 and world TuMV isolates at nucleotide level. Phylogenetic relationship was determined among CK01 and TuMV isolates from different parts of the world. To our knowledge; although TuMV infections were identified in different plants, this is the first report of TuMV infection on canola plants in Turkey.
