Interaction with its ligand, stem cell factor, is essential in the development of many cells in human body. Especially in the hematopoietic stem cells could be protected from apoptosis as a result of KIT receptor activation which up-regulates bcl-2 expression. This study examines the ratio of C-kit expression in uterin leiomyosarcomas (LMS) and determines the correlation between C-kit expression and bcl-2 up-regulation. Materials and Methods: Archival tissue blocks from 10 patients with LMS and 10 leiomyoma (LM), which were randomly selected, were retrieved from pathology department. Normal myometrial tissues from patients with LM were determined and third group was constituted. Tissue samples were stained with monoclonal mouse antibody for bcl-2 and polyclonal rabbit antibody for Ckit. Avidin biotin immunoperoxidase method was employed for antibody detection. Staining intensity was graded 1+, 2+ or 3+ for bcl-2 and C-kit, staining distribution was graded as focal (10-30%), intermediate (30-60%) or diffuse (>60%) for C-kit. Results: C-kit was positive in 6 (%60), 1 (%10) and 0 (%0) of the LMS, LM and myometrium respectively.
