Bu amaçla, optimizasyon işlemi içintepki yüzey yöntemi tabanlı merkezi tümleşik tasarım dizaynındanfaydalanılmıştır. Örneklerin hazırlanması amacıyla sıvı/katı oranı ve etanolkonsantrasyonu faktör olarak seçilmiştir. Örneklerin biyoaktif özelliklerininbelirlenmesi amacıyla toplam fenolik madde miktarı, antiradikal kapasite ve EC50değeri ile verim özellikleri hesaplanmıştır. Sıvı/katı oranı ve etanolkonsantrasyonunun örneklerin toplam fenolik madde miktarı üzerinde oldukça etkiliolduğu görülmüştür. Örneklerin toplam fenolik madde miktarı değerleri 605-1754mg GAE/L aralığında, EC50 değerleri 9.80-199.2 aralığında, ekstraktverimleri ise %2.32-6.49 aralığında değişim göstermiştir. Bitki örneklerinde sıvımiktarının artışı toplam fenolik madde miktarını artırırken ekstrakt verimindedüşüşe yol açmıştır. Optimizasyon işlemi sonucunda en yüksek toplam fenolikmadde miktarının %57 etanol konsantrasyonunda ve 1:4 katı/sıvı oranında eldeedilebileceği sonucuna ulaşılmıştır. KW - Karahindiba KW - tentür KW - biyoaktivite KW - optimizasyon KW - tepki yüzey yöntemi N2 - In this study, dandelion (Taraxacumofficinale) plant was used for the production of tincture, a hydroalcoholicextract type, and it was aimed to determine process factor levels for theoptimization of the production conditions in order to increase the final productquality. For this purpose, central composite rotatable design based on responsesurface methodology was utilized for the optimization process. The liquid /solid ratio and the ethanol concentration were chosen as factors for thepreparation of the samples. In order to determine bioactive properties of samples,total phenolic content, antiradical capacity (EC50) value andextract yield properties were determined. Liquid / solid ratio and ethanolconcentration were found to be very effective over the total amount of phenolicmaterials. The total phenolic content values ​​of the samples ranged from605-1754 mg GAE/L, EC50 values ​​ranged from 9.80-199.2, and extractyields ranged from 2.32-6.49%. Increasing the amount of liquid for herbsincreased the total phenolic content, which led to a decrease in the yield ofthe extract. The result of the optimization process showed that the highesttotal phenolic content could be achieved in a 1: 4 solid / liquid ratio at aconcentration of 57% ethanol.
