In the present study, the mycotoxin citrinin (CTN) was evaluated for their effects on mitotic index (MI) rates of human peripheral blood lymphocytes. Heparinized peripheral blood samples were obtained from six healthy non-smoking individuals (3 females and 3 males) and cultured for 72 h in the culture medium Nutrient Mixture F-10 in 9 different tubes for each individual. Lymphocyte cultures were treated for the last 48 h with CTN at the following concentrations, 10 mM, 20 mM, 40 mM, 60 mM, 80 mM, 100 mM, and 0.1 mM mitomycin c (positive control) and 0.5% absolute ethanol. To block metaphases, at 70 h of incubation, colchicine was added to cultures to give a final concentration of 0.1 μg/ml, and cultures were harvested cells at 72 h. At 60 mM, 80 mM and 100 mM CTN concentrations, significantly reduced MI values in comparison with negative controls were found (p=0.001). In addition, CTN caused a significant concentration-dependent decrease in MI rates in human lymphocytes.
