Misellar sıvı kromatografisinde kullanılan yüzey aktif maddelerin konsantrasyonu kritik misel konsantrasyonunu aştığında taşıyıcı fazda miseller oluşmaya başlar. Kullanılan yüzey aktif madde ve taşıyıcı fazın özelliklerine konsantrasyon, iyonik güç, sıcaklık, basınç bağlı olarak oluşan miseller de farklıdır. En yaygın olarak kullanılan yüzey aktif madde sodyum dodesil sülfattır. Misellar taşıyıcı faz düşük toksisite, düşük maliyet, aynı analitik çalışmada iyonik ve iyonik olmayan analitlerin eşzamanlı belirlenmesini sağlama ve çevreye olan olumsuz etkilerin azaltılması gibi bazı avantajlara sahiptir. En önemli dezavantajlarından biri ise taşıyıcı fazın kromatografik sistemden sürekli akıtılmak zorunda olunmasıdır. Günlük kolon temizliği ve kolon şartlandırılması ters faz sıvı kromatografisine kıyasla daha çok zaman alır ve bazen kolon zarar görebilir. Misellar kromatografi gıda analizlerinde yaygın olarak kullanılmaktadır. In MLC, surfactant concentrations in excess of the critical micelle concentration are used so that micelles are formed in the mobile phase. Depending on the type of surfactant and properties of the mobile phase concentration, ionic strength, temperature, and pressure , the shape of micelles is different. The most commonly used surfactant in MLC is sodium dodecyl sulfate SDS . Micellar phase has some advantages over other phases, including low cost, low toxicity, the possibility of separation of ionic and non-ionic compounds on the same analytical run and the reduced environmental impact. One of the main disadvantages of the micellar chromatography, the mobile phase should be continuously flushed through the chromatographic system. Daily cleaning and re-equilibration of column takes more time compared to reverse-phase chromatography and sometimes, column may be damaged. Micellar chromatography is widely used in food analyses.
