Liriomyza sativae Blanchard, 1938 (Diptera: Agromyzidae)’nin DNA Barkodlaması

Abstract

Bu çalışmada İncirliova ve Koçarlı ilçelerinde (Aydın) sebze alanlarından toplanmış Liriomyza sativae’ nin DNA barkodlaması 2020-2022 yılları arasında yapılmıştır. Morfolojik olarak tür teşhisi yapıldıktan sonra; örneklerin COI gen bölgesinin çoğaltılması için HCO/ LCO evrensel primerler kullanılmış ve 612 bç COI bölgesi elde edilmiştir. Örneklerin sekans analizi Triogen Biyoteknoloji (İstanbul-Türkiye) tarafından yapılmıştır. Örnekler NCBI’da bulunan BLAST aracılığıyla kontrol edildiğinde türlerin L. sativae (% 99-100) ve L. trifolii (%97-99) olduğu görülmüştür. Örnekler Genbank’a kaydedilmiştir. Çalışma sonucunda genetik çeşitlilik düşük bulunmuştur.

DNA Barcoding of Liriomyza Sativae Blanchard Agromyzidae, 1938 (Diptera: Agromyzidae

Abstract

In this study, DNA barcoding of Liriomyza sativae collected from vegetable areas in Incirliova and Koçarlı districts (Aydın) was performed between 2020-2022. After the morphological diagnosis of L. sativae, HCO/LCO universal primers were used to reproduce the COI gene region of the samples and 612 bp COI regions were obtained. The sequence analysis of the samples was performed by Triogen Biotechnology (Istanbul, Türkiye). When the samples were checked through the BLAST located at NCBI, it was found that the species were L. sativae (99-100%) and L. trifolii (97-99%). The samples were registered to Genbank. As a result of the study, genetic diversity was found to be low.
