The Effects of Achillea millefolium (Yarrow) Extract and Luteolin on In Vitro Rat Urinary Bladder Muscles

Abstract

Yarrow (CP) is commonly used in alternative medicine and in its structure is found Luteolin together with many

compounds. In this study, effects of CP methanol extract and Luteolin on isolated rat bladder smooth muscle contractility

were investigated. In the study, twenty male Sprague Dawley rats were used and the rats were decapitated under anesthesia.

The cleaning of isolated bladders were done and muscle strips prepared from the bladders were placed to organ baths. The

tissues were applied to 1 gram tesnion and left for 1 hour incubation period. At the end of the incubation period, CP extract,

Luteolin, acetylcholine (ACh), potassium chloride (KCl) and Nifedipine was applied with a protocol the bath. Data were

evaluated by Tukey test in one way ANOVA. It was determined that different doses of CP extract and Luteolin inhibited the

isolated bladder contractility induced by ACh and KCl. It was seen that of the contractions induced by ACh submaximal dose

more strongly suppressed by CP extract and Luteolin in the presence of Nifedipine in the bath. Subcutaneous dose-induced

indications of KCl were found to be more inhibitory in co-existence of CP extract and Nifedipine. It was determined that

together with CP extract and Nifedipine were more inhibited the KCl’s submaximal dose-induced contractions, but but the

effect of Luteolin in the presence of Nifedipine in bath was not changed. In this study, it was assigned that inhibiting the

bladder smooth muscle contractions of different doses of CP extract and Luteolin and of CP extract to be also effective on the

M3 receptor pathway.

Achillea millefolium (Civanperçemi) Ekstraktı ve Luteolin’in İn Vitro Rat Mesanesi Düz Kas Kontraksiyonları Üzerine Etkileri

Abstract

Civanperçemi (CP) alternatif tıpta yaygın olarak kullanılmakta ve yapısında birçok bileşikle birlikte Luteolin de

bulunmaktadır. Bu çalışmada, CP metanol ekstraktı ve Luteolin’in izole rat mesane düz kas kontraktilitesi üzerine etkileri

araştırıldı. Çalışmada 20 adet erkek Sprague Dawley rat kullanıldı ve ratlar anestezi altında dekapite edildiler. İzole edilen

mesanelerin temizlikleri yapıldı ve mesanelerden hazırlanan kas şeritleri organ banyosuna asıldı. Dokuya 1 gram gerim

uygulandı ve 1 saatlik inkübasyon periyoduna bırakıldı. İnkübasyon periyodu sonunda banyoya CP ekstraktı, Luteolin,

asetilkolin (ACh), potasyum klorür (KCl) ve Nifedipin bir protokol çerçevesinde uygulandı. Veriler one way ANOVA’da Tukey

testi ile değerlendirildi. CP ekstraktı ve Luteolin’nin farklı dozlarının, ACh ve KCl ile indüklenen izole mesane kontraksiyonlarını

inhibe ettikleri belirlendi. ACh’nın submaksimal dozu ile indüklenen kontraksiyonların banyoda Nifedipin varlığında CP

ekstraktı ve Luteolin ile daha güçlü bir şekilde baskılandığı görüldü. KCl’nin submaksimal dozu ile indüklenen kontaksiyonları

CP ekstraktı ile Nifedipin’in birlikte daha fazla inhibe ettikleri belirlendi, ancak banyoda Nifedipin varlığında Luteolin’in

etkisinin değişmediği tespit edildi. Bu çalışmada CP ekstraktı ve Luteolin’nin farklı dozlarının mesane düz kas kontraksiyonlarını

inhibe ettikleri ve CP ekstraktı ve Luteolin’in L tipi Ca2+ kanalları aracılığıyla etkili olurken, CP ekstraktının ayrıca M3 reseptör

yolağı üzerinden de etkili olabileceği belirlendi.
