                            <p>This clinical study evaluated the antihypertensive efficacy and side effects of DCR 515 (Briserin) in 27 patients with predominantly essential hypertension. The compound combines dihydroergocrystin, reserpine, and clopamide. Patients received initial doses of 1–2 tablets daily, followed by adjustments. Over a 4-week period, average systolic BP decreased from 202 mmHg to 140 mmHg (31% reduction) and diastolic BP from 117 mmHg to 86 mmHg (26% reduction), both statistically significant. Side effects such as fatigue, dizziness, nausea, and appetite loss occurred in a few patients, but were reversible upon dose reduction or withdrawal. Serum potassium decreased slightly (from 4.6 to 3.8 mEq/L), yet changes were not statistically significant. In 6 cases, transient increase in urea was noted, with one chronic nephritis patient showing persistent elevation. No significant EKG, retinal, or hepatic changes were observed. Authors conclude DCR 515 is effective in reducing BP, especially diastolic levels, with manageable side effects in compliant patients.</p></trans-abstract>
                                                                                                                                    <abstract><p>Bu klinik çalışmada, DCR 515 (Briserin) bileşiğinin 27 hastada hipertansiyon üzerindeki etkinliği ve yan etkileri değerlendirilmiştir. Bileşim; dihidroergokrystin, reserpin ve klopamid içerir. Hastalara günlük 1–2 tablet ile tedavi başlanmış, doz kişisel tepkiye göre ayarlanmıştır. Dört haftalık takipte, ortalama sistolik tansiyon %31 oranında 202 mmHg&#039;den 140 mmHg&#039;ye; diyastolik tansiyon ise %26 oranında 117 mmHg&#039;den 86 mmHg&#039;ye düşmüştür. Her iki değer istatistiksel olarak anlamlı bulunmuştur. Yorgunluk, baş dönmesi, bulantı, iştahsızlık gibi yan etkiler 6 vakada görülmüş; doz azaltımı veya tedavinin kesilmesiyle kaybolmuştur. Serum potasyum değerleri (%21 azalma) klinik olarak anlamlı düşmemiştir. 6 vakada üre düzeyinde geçici artış gözlenmiş; bir kronik nefritli hastada ise bu yükseklik kalıcı olmuştur. EKG, retinopati veya karaciğer fonksiyonlarında değişiklik izlenmemiştir. Araştırmacılar, Briserin’in özellikle diyastolik basıncı düşürmede etkili ve tolere edilebilir bir seçenek olduğunu belirtmiştir.</p></abstract>
