17-β Estradiole ile İndüklenen insan Lenfositlerindeki Mikronükleus Sıklığının incelenmesi

Abstract

Bazı epidemiyolojik ve deneysel çalışmalar, kanser gelişimi ile östrojenler arasında bir ilişkinin olduğunu göstermiştir. Estrojenler ile ilişkili karsinogenezin, reaktif metabolitlerle direk DNA hasarı ve reaktif oksijen metabolitleriyle indirek genotoksik etkinin bir kombinasyonunun sonucu olduğunu düşündürmektedir. İnsan periferal kan lenfositlerinde uygulanan 'Sitokinez- blok' mikronükleus (MN) metodu, genetik toksikoloji testleri arasında standart bir sitogenetik test olarak dünya üzerindeki birçok laboratuarca kabul edilen bir metodtur. Sunulan çalışmanın amacı, lenfosit kültürlerinde estradiol (E2)'ün muhtemel genotoksik etkilerini 'Sitokinez-blok' MN metodu kullanarak araştırmaktır. Çalışma, 50 sağlıklı erkek donörden izole edilen lenfositlerde gerçekleştirildi. Kontrol kültürleriyle (1-15 MN / 1000 hücre) karşılaştırıldığın da E2 (36 μM) uygulanan kültürlerde (1-11 MN / 1000 hücre) MN sıklığında herhangi bir istatistiksel fark tespit edilmedi (p > 0.05). Ayrıca binükleuslu hücredeki MN sıklığı, E2 uygulaması ile değişmedi (Kontrol kültürlerinde: 5.88 ± 2.9; E2 kültürlerinde 5.64 ± 2.5, p > 0.05). Sonuçlarımız; estradiolün kültüre edilmiş lenfositlerde herhangi bir genotoksik etkisinin olmadığını göstermiştir.
