The first record and barcoding of the Mediterranean fruit fly Ceratitis capitata (Diptera: Tephritidae) in Kırşehir, Turkey

Abstract

The Mediterranean fruit fly Ceratitis capitata (Wiedemann) (Diptera: Tephritidae) is a cosmopolitan fruit pest occuring in the Mediterranean, Aegean and Black Sea regions of Turkey. Although it stays more limited and less dangerous, its occurrence in localized favorable habitats in Central Anatolia (Ankara) has been known for a long time. Here we provide the first record of the pest in Kırşehir (Central Anatolia). We conducted fruit (peach, pear and plum) sampling in the region and identified the emerging specimens through both morphology and molecular barcoding (partial mtCOI). We propose two hypotheses to explain presence of the medfly in Kırşehir. First, it may enter Kırşehir via fruits transported for commercial purposes from cities in southern Turkey and it may establish temporary summer populations in Kırşehir where winter temperatures are not favorable for its survival. Second, it may somehow establish permanent populations surviving low winter temperatures in Kırşehir. In order to test the latter, we placed fruits infected with the medfly, found in Kırşehir, on the ground in a fruit garden in Kırşehir and covered it with a net to protect from predation and to catch adults. We could not find any adult in the net until the following spring. This observation does not support the second hypothesis but sophisticated overwintering experiments should be conducted. Moreover, the second hypothesis can still be valid since the medfly can overwinter indoors. This study is the first record of C. capitata in Kırşehir. We also provide the first medfly mitochondrial sequence from Turkey.

Akdeniz meyve sineği Ceratitis capitata (Diptera: Tephritidae)’nın Kırşehir’deki ilk kaydı ve barkodlaması

Abstract

Akdeniz meyve sineği, Ceratitis capitata (Wiedemann) (Diptera:
Tephritidae) kozmopolit bir meyve zararlısıdır. Türkiye’de Akdeniz, Ege ve
Karadeniz Bölgesi’nde yayılış gösterdiği bilinmektedir. İç Anadolu’da büyük
zararlara neden olmamakla birlikte, yaşamasına elverişli sınırlı habitatlardaki
varlığı uzun zaman önce Ankara’da tespit edilmiştir. Bu çalışma kapsamında
soğuk kış koşullarının yaşandığı Kırşehir’de gerçekleştirilen armut, şeftali ve
erik örneklemelerinde de
C. capitata
tespit edilmiştir. Zararlının tespiti hem morfolojik, hem de mitokondriyel COI
gen bölgesine ait primer ile moleküler barkodlama yapılarak gösterilmiştir. Bu
çalışmada, Kırşehir’de bilinen biyolojisine uygun olmamasına karşın,
C. capitata’nın görülmesi ile ilgili
olarak iki hipotez öne sürülmüştür. Birincisi
C. capitata’nın Kırşehir’e, şehir dışından satılmak üzere getirilen
meyvelerle geldiği, Kırşehir’de geçici yaz populasyonları oluşturduğu ancak
soğuk geçen kışlar nedeniyle burada kışlamadığıdır. İkincisi ise
C. capitata’nın Kırşehir kış şartlarında
hayatta kalabilen yerleşik bir populasyonunun olduğudur. İkinci hipotezi
sınamak için bu çalışma kapsamında, Kırşehir’de yapılan örneklemelerden elde
edilen
C. capitata larvası bulunan
meyveler bir meyve bahçesinde toprak üzerine yerleştirilmiş ve üzeri,
predasyondan korumak ve çıkan erginleri yakalamak amacıyla sinek teli ile
kapatılmıştır. İzleyen ilkbahara kadar yapılan gözlemlerde burada herhangi bir
ergine rastlanmamıştır. Bu sonuç ikinci hipoteze ilişkin olumsuz bir kanıttır.
Ancak bunun sağlıklı bir şekilde ortaya konması için daha kapsamlı kış
denemelerinin yapılması gerekmektedir. Bununla birlikte
C. capitata ev ya da ahır gibi kapalı mekanlarda
kışlayabileceğinden ikinci hipotezin hala geçerli olabileceği göz ardı
edilmemelidir. Bu çalışma
C. capitata’nın
Kırşehir’deki ilk kaydıdır. Ayrıca bu çalışma ile Türkiye’den ilk kez
C. capitata mitokondriyel dizi verisi de
bildirilmektedir.
