                                                                                                <abstract><p>Amaç: Temporomandibular EklemDüzensizliği (TME-İD) ve eklem osteoartritine (TME-OA) yatkınlık genetikvaryasyonlarla ilişkili olabilir. Vitamin D Reseptörü (VDR) gen polimorfizmleri buna adaydırlar.Bu çalışmanınamacı, VDR FokI polimorfizminintemporomandibulareklem dejenerasyonu ile ilişkili olup olmadığınıcinsiyete göre değerlendirmekti. Yöntem: 58 osteoartritikTME-İD hastası (32.07.±8.1) ve kontrol olarak 71 TME-İD olmayan birey(28.28±5.95)çalışmaya dahil edildi. Kan örneklerindenDNA izolasyonu standart proteinazK/fenol-kloroformmetodu ile yapıldı. VDR geni FokI polimorfizmi, polimeraz zincir reaksiyonunu (PZR) takiben restriksiyonfragment uzunlukpolimorfizmi(RFLP) yöntemi ilearaştırıldı. Bulgular:FokI genotipdağılımları(rs2228570, C&amp;gt;T) grupları arasında istatistiksel olarakfarklı idi (p=0.026, χ²=7.2). Heterozigot Ff genotipi, FF genotipine göre istatistiksel olarak farklı idi (OR=0.43, %95 GA:0.2-0.92, p=0.028). TME-İD’li kadınlarda heterozigot Ffgenotipi FF genotipine göre farklılık sınırında idi (OR=0.43, %95 CI:0.16-1.10, p=0.07). Yine TME-İD’li kadınlarda ff genotipiile FF genotipi arasında fark olmamasına rağmen (%95 GA: 0.29-26.03, p=0.37) risk faktörü 2.77 kat fazla idi. TME-İD/kontrol grubu ve kadın kontrollerde de F ve f alellerinin dağılımları farklı değildi. Sonuç: FokI polimorfizmininTME-İD/TMEOA gelişmesi üzerinde önemli bir etkisi olduğu düşünülebilir. ffgenotipi, TME-İD hastalarında ve TME-İD’li kadınlarda temporomandibular eklemdejenerasyonu ile ilgili olabilir.</p></abstract>
                                                                                                                                    <trans-abstract xml:lang="en">
                            <p>Objective: Predisposition to temporomandibular jointinternalderangement (TMJ-ID) and osteoarthritisof the joint(TMJOA)might be related to genetic variations.Vitamin D receptor (VDR) genepolymorphismsare the candidates forassociation with the disorder. The aim ofthis study was to evaluatewhether VDRFokI polymorphism is associatedwith the degeneration of the temporomandibular jointbygender.Methods: The study included 58 unrelated TMJ-ID patients(32.07±8.1) and 71 healthy controls (28.28 ±5.95) without TMJ-ID. DNA extractionwas achieved by standardproteinaseK/phenol-chloroformmethodfrom the blood samples.VDR gene FokI polymorphism was investigatedby a polymerasechain reaction(PCR) based restrictionfragment length polymorphism(RFLP) assay.Results: The genotype distributionsof FokI (rs2228570, C&amp;gt;T) showed statisticallysignificant results (p=0.026,χ²=7.2). Ff heterozygote genotype was statistically different as compared to FF genotype (OR=0.43, 95% CI: 0.2-0.92,p=0.028).HeterozygoteFf genotypewas differentcomparing to the FFgenotype in TMJ-ID women (OR=0.43, 95% CI: 0.16-1.10,p=0.07).Although the difference between ff and FF genotypewere not different, ff genotype was a 2.77 foldrisk factor(95% CI:0.29-26.03, p=0.37).The distributions of F and f alleles were not different between the groups ofoverall TMJ-ID vs. controls and women TMJ-ID vs. controls. Conclusion: The results of this study suggested that FokI polymorphism mightpresent susceptibility to TMJ-ID/TMJOA. ff genotype mightbe associatedwith the degeneration of temporomandibular joint in TMJ-ID patients and TMJ-ID women.</p></trans-abstract>
