GLUKOZ 6-FOSFAT DEHIDROGENAZ ÜZERİNE BAZI SİTOTOKSİK KİMYASALLARIN ETKİSİ

Abstract

Özet: Bu çalışmanın amacı, glukoz 6-fosfat dehidrogenaz (G6PD) üzerine bazı sitotoksik kimyasalların in vitro etkilerini araştırmaktır. Başlangıçta, G6PD enzimi insan eritrositlerinden amonyum sülfat çöktürmesi ile kısmi olarak ve bunu takiben afinite kromatografisi ile saflaştırıldı. Inhibisyon etkisi gösteren ilaçların I50 değerleri %aktivite - ilaç konsantrasyonu grafiği ile belirlendi. Saflaştırılan enzim üzerinde araştırılan iki ilacın (2,4-dihidroksi-5-floropirimidin ve sodyum 2-sulfaniletansulfonat) enzimi inhibe ettiği belirlendi.

EFFECT OF SOME CYTOTOXIC CHEMICALS ON GLUCOSE 6-PHOSPHATE DEHYDROGENASE

Abstract: The objective of this study was to determinate in vitro effects of some cytotoxic chemicals (2,4-dihydroxide-5-fluoropirimidine and sodium 2-sulfanylethanesulfonate) on glucose–6-phosphate dehydrogenase (G6PD). Initially, G6PD was purified from human erythrocyte by using ammonium sulphate precipitation and affinity chromatography. I50 values of the drugs exhibiting inhibition effects were determined by means of [drug]-activity% graph. The drugs investigated on the purified enzyme inhibited the enzyme activity.
