Özet

Amaç: BRCA1 ve BRCA2 genlerindeki mutasyonlar, kalıtımsal meme kanserlerinde predispozan faktördür. Sporadik meme kanserlerinde de BRCA proteini ekpresyonunda kayıp olduğu görülmektedir. Bu çalışmada amacımız, Isparta'da meme kanserlerinde BRCA ekspresyonunu değerlendirmek ve prognostik faktörlerle korelasyonunu araştırmaktır. Materyal ve metod: 82 seçilmemiş meme kanserinden, formalinde fiske edilmiş ve parafine gömülmüş doku kesitlerine BRCA1 ve BRCA2 immunohistokimyasal boyaması yapıldı. Sonuç: 82 meme kanserli hastada 39(%47.57) BRCA1 ve 42(%50.12) BRCA2 ekspresyonunu pozitif bulduk. 40(% 48.77) karsinomda BRCA ekspresyonu kayıp idi. Bu çalışmanın sonucunda BRCA proteinleri ekpresyonlarının immunhistokimyasal olarak tespiti ile familyal olan ve olmayan meme kanserlerinin birbirinden ayrılamayacağı, ancak histolojik ve nükleer grade yükseldikçe BRCA protein ekpresyonlarının da azalması, BRCA nın diferansiyasyon belirleyici bir marker olarak kullanılabileceğini düşündürdü.

Abstract



BRCA1 and BRCA2 expression in breast cancer patients in ısparta

Aim: Mutations in BRCA1 and BRCA2 genes predispose to hereditary breast cancer. Loss of expression of BRCA proteins have been seen in sporadic breast carcinomas. Our aim in this study, was to evaluate, expression of BRCA and its correlation with prognostic factors. Material and method: Immunohistochemical staining for BRCA1 and BRCA2 was performed on sections formalin- fixed and parafin-embedded tissue from 82 unselected invazive breast carcinoma. Result: We found 39 (47.57%) BRCA1 and 42 (50.12%) BRCA2 positive in 82 breast carcinomas. Expression of BRCA was lost in 40(48.77 %) carcinomas. At the end of this study, with immunohistochemical evaluation of expression of BRCA proteins can not differantiate familial and nonfamilial breast carcinomas. As histological and nuclear grades are increased, expression of BRCA protein is decreased, therefore expression of BRCA can be used as a differentiation parameter.
