Çorum Yöresinde İnsanlar Üzerinde Parazitlenen Kenelerde Riketsiya Varlığının Araştırılması

Abstract

Amaç: Bu çalışmada, Çorum yöresinde insanlarda parazitlenen kenelerde riketsiya varlığının Polimeraz Zincir Reaksiyonu PZR yöntemiyle araştırılması amaçlanmıştır.Yöntem: Çorum yöresinde insanlar üzerinden toplanan 1.010 adet kene toplanarak morfolojik karakterlerine göre tür teşhisleri yapılmıştır. Bu örneklerden bireysel olarak elde edilen total DNA’lar riketsiyal sitrat sentaz gltA, 381 bp ve dış membran protein A ompA, 532 bp gen bölgelerini hedefleyen primer setleri kullanılarak PZR yöntemi ile taranmıştır.Bulgular: Çorum ilinde insanlar üzerinden toplanan 741 Hyalomma marginatum örneğinin 51 %6,88 ’inde Rickettsia aeschlimannii, 3 %0,4 ’ünde Rickettsia sibirica mongolitimonae; 32 Dermacentor marginatus raoultii, 3 %9,4 ’ünde Rickettsia slovaca varlığı tespit edilmiştir. Çalışma kapsamında incelenen Hyalomma aegyptium, Hyalomma excavatum, Hyalomma parva, Haemaphysalis punctata, Ixodes ricinus, Rhipicephalus bursa ve Rhipicephalus turanicus türlerine ait kenelerde riketsiyal DNA varlığına rastlanmamıştır.Sonuç: Bu çalışmada, Çorum yöresindeki kenelerde

Investigation of the Presence of Rickettsiae in Ticks Parasitizing Humans in Corum Region

Abstract

Objective: The aim of this study is to determine the rickettsiae in ticks collected on human in Çorum Province by using the Polymerase Chain Reaction PCR .Methods: A total of 1010 tick samples which were collected on humans identified to species level in terms of the morphological characters. Total DNA’s individually extracted from ticks were screened for the presence of Spotted Fever Group rickettsiae using the PCR targeting rickettsial citrate synthase gltA, 381 bp and outer membrane protein ompA, 532 bp genes.Results: Out of 741 Hyalomma marginatum ticks collected from humans in Çorum Province, 51 6.88% were infected Rickettsia aeschlimannii, 3 0.4% were infected Rickettsia sibirica mongolitimonae. Out of 32 Dermacentor marginatus ticks, 3 9.4% were infected Rickettsia raoultii and 3 9.4% were infected Rickettsia slovaca. In addition, rickettsial DNA was not found in Hyalomma aegyptium, Hyalomma excavatum, Hyalomma parva, Haemaphysalis punctata, Ixodes ricinus, Rhipicephalus bursa and Rhipicephalus turanicus ticks.Conclusion: In the present study, rickettsiae in ticks collected from Çorum Province were examined GIRIŞ Bilinen en eski vektör kaynaklı hastalık ajanlarından
