Bu patojenler çoğunlukla yoğun bakım hastalarından izole edilirler. Hastalar, bir kişiden diğerine hastalığın bulaşmasında birincil kaynak olmakla birlikte, tıbbi ekipman ve hastane personeli de enfeksiyonun yayılmasından sorumludur. Bu tez çalışmasında, klinik örneklerden elde edilen Acinetobacter suşları üzerinde PCR yöntemini uygulayarak plazmit aracılı kinolon direnç genleri araştırılmış ve böylece bu genlerin varlığının ve yaygınlığının belirlenmesiyle birlikte ve Acinetobacter türlerinin kinolon direncinde plazmitlerin rolünün daha iyi anlaşılması beklenmektedir. Materyal ve Metot: Ocak 2021 - Temmuz 2021 tarihleri arasında Tıbbi Mikrobiyoloji Laboratuvarında rutin tanı amacıyla gönderilen klinik örneklerden 175 Acinetobacter izolatı çalışmaya dahil edilmiştir. Daha sonra, Acinetobacter izolatlarından DNA ekstraksiyonu gerçekleştirilmiş ve ardından multipleks PCR yöntemi kullanılarak kinolon direnç genlerinin varlığı araştırılmıştır.Bulgular: Bu araştırmanın birincil amacı, acinetobacter türlerine karşı kinolon ilaç etkinliğini önleyen direnç genlerinin varlığını belirlemektedir. Klinik laboratuvar rutininden elde edilen toplam 175 örneği Acinetobacter türlerinde gen direncini belirlemek için kullanılmıştır. Bulgularımıza göre qnr-S geninin diğer genlere göre daha baskın olduğu gözlenmiştir. qnr A, qnrB, qnrC, qnrS, oqxAB, oqxA ve qepA genlerinin direnç oranı sırasıyla %0.57, %0.57, %0.57, %5.71, %2.8, %4, %0 şeklinde olduğu belirlenmiştir.Sonuç: Çalışmamızda plazmit aracılı kinolon direnç genleri A.baumannii izolatlarında tespit edilmiştir. Bu konuda ülkemizde çok fazla yayın bulunmamaktadır. KW - Acinetobacter baumannii KW - kinolon KW - direnç KW - qnr KW - oqxAB N2 - Objective: Acinetobacter isolates are opportunistic nosocomial pathogens commonly found in hospital settings. These pathogens are mostly isolated from intensive care patients. Although patients are the primary source of transmission from one person to another, medical equipment and hospital staff are also responsible for the spread of infection. In this thesis, we investigated plasmid-mediated quinolone resistance genes by PCR on Acinetobacter strains obtained from clinical specimens. By determining the presence and prevalence of these genes, we expect to better understand the role of plasmids in quinolone resistance of Acinetobacter species. Materials and Methods: Between January 2021 and July 2021, 175 Acinetobacter isolates from clinical specimens sent for routine diagnosis in the Medical Microbiology Laboratory were included in the study. Subsequently, DNA extraction was performed from Acinetobacter isolates and then the presence of quinolone resistance genes was investigated using multiplex PCR method.Results: The primary objective of this study was to determine the presence of resistance genes that prevent quinolone drug efficacy against Acinetobacter species. A total of 175 samples obtained from clinical laboratory routine were used to determine gene resistance in Acinetobacter species. According to our findings, qnr-S gene was observed to be more dominant than other genes. The resistance rates of qnr A, qnrB, qnrC, qnrS, oqxAB, oqxA and qepA genes were 0.57%, 0.57%, 0.57%, 5.71%, 2.8%, 4% and 0%, respectively.Conclusion: In our study, plasmid-mediated quinolone resistance genes were detected in A. baumannii isolates.
