Bu çalışmada, Anadolu'dan dört Centaurea türünün antiaging ve antidiyabet etkilerini belirlemesi amaçlanmaktadır. Ayrıca Centaurea lycopifolia, C. balsamita, C. depressa, ve C. iberica türlerinin GC-MS ile bazı triterpen içerikleri belirlenmiştir. C. lycopifolia türünün aseton ekstresinde (4271.54 μg/g ekstre), C. balsamita türünün aseton ekstresinde (2872.28) ve metanol ekstresinde (3564.05) α-amirin tespit edilmiştir. Antiaging açısından elastaz ve kollajenaz yöntemlerinde C. lycopifolia aseton ekstresinin elastaz enzimine karşı iyi derecede (27.83±1.31, oleanolik asit:37.47±0.82) inhibe edici aktivite sergilemiştir. Antidiyabetik açısından değerlendirdiğimizde ise C. depressa türünün aseton ekstresinin 12.5, 50 ve 200 µg/mL farklı konsantrasyonlarda α-glukozidazı çok yüksek düzeyde (sırasıyla, % inhibisyon: 29.03±0.49, 45.87±1.06, 84.81±1.22, akarboz: 1.23±0.03, 28.51±0.44, 64.37±1.80) inhibisyon aktivitesi gösterdiği belirlenmiştir. C. depressa türünün metanol ekstresi ile C. iberica ve C. lycopifolia türlerinin aseton ekstreleriinin α-glukozidaz enzimini yüksek düzeyde inhibe ettiği belirlenmiştir. C. balsamita türünün metanol ekstresinin ise 800 µg/mL konsantrasyonda α-amilaz enzimine karşı yüksek düzeyde (65.43±1.40, akarboz: 67.18±1.73) inhibisyon aktivitesi gösterdiği tespit edilmiştir. Sonuç olarak C. lycopifolia türünün antiaging potansiyeli nedeniyle kozmetik endüstrisinde, C. depressa, C. iberica, C. lycopifolia ve C. balsamita türlerinin antidiyabetik potansiyelleri nedeniyle farmasötik alanda kullanılabilmesi için daha detaylı çalışmalara konu olması gerektiği düşünülmektedir. KW - Centaurea KW - GC-MS KW - α-amirin KW - enzim aktivite KW - antiaging KW - antidiyabetik N2 - Centaurea L. is the fourth largest genus of the Asteraceae family, with more than 600 species distributed worldwide, especially in Western Asia and the Mediterranean regions, and there are 179 species in Turkey, 109 of which are endemic. This study aims to determine the antiaging and antidiabetes effects of four Centaurea species from Anatolia. Additionally, some triterpene contents of Centaurea lycopifolia, C. balsamita, C. depressa, and C. iberica species were determined by GC-MS. α-amyrin was detected in the acetone extract of C. lycopifolia species (4271.54 μg/g extract), in the acetone extract of C. balsamita species (2872.28) and in the methanol extract (3564.05). In terms of antiaging, elastase and collagenase methods; C. lycopifolia acetone extract exhibited good (27.83±1.31, oleanolic acid:37.47±0.82) inhibitory activity against the elastase enzyme. When we evaluate it from an antidiabetic perspective, the acetone extract of the C. depressa species has very high levels of α-glucosidase at different concentrations of 12.5, 50 and 200 µg/mL (respectively, % inhibition, 29.03±0.49, 45.87±1.06, 84.81±1.22, acarbose: 1.23±0.03, 28.51±0.44, 64.37±1.80) was determined to show inhibition activity. It was determined that the methanol extract of C. depressa species and the acetone extracts of C. iberica and C. lycopifolia species inhibited the α-glucosidase enzyme at a high level. It was determined that the methanol extract of C. balsamita species showed high level (65.43±1.40, acarbose: 67.18±1.73) inhibition activity against the α-amylase enzyme at a concentration of 800 µg/mL. As a result, in the cosmetic industry due to the antiaging potential of C. lycopifolia species, it is thought that C. depressa, C. iberica, C. lycopifolia and C. balsamita species should be subject to more detailed studies in order to be used in the pharmaceutical field due to their antidiabetic potential.
