Gemlik, Sarı Haşebi, Halhalı (Hatay), Saurani ve Kilis Yağlık zeytin çeşitleri 1 yaşındaki çelikten üretilmiş Gemlik anacına yama aşı yöntemi ile aşılanmıştır. 1 yaşına gelen aşılı fidanların floem dokularından örnekler alınarak etüvde 65 oC de kurutulmuş ve porselen havanda öğütülerek analize hazır hale getirilmiştir. Ardından 1 gr örneğe 10 ml metanol eklenerek hazırlanan süzüntü HPLC ye enjekte edilerek analizler yapılmıştır. Zeytinde yoğun olarak bulunan fenolik bileşiklerin (protokateşik asit, kafeik asit, p-kumarik asit, ferulik asit, rutin trihidrat ve kuersetin) bulunma durumları belirlenmiştir. Çalışma sonuçları incelendiğinde; protokateşik asit, kafeik asit, rutin trihidrat ve kuersetin aşı noktasının üzerinde yüksek düzeyde tespit edilirken p-kumarik asit, ferulik asit aşı noktasının altında daha yüksek düzeyde belirlenmiştir. Aşı kaleminde yüksek oranda bulunmasının uyuşmazlıkla sonuçlandığı değerlendirilen fenolik bileşikler (ferulik asit ve p-kumarik asit) tüm kombinasyonlarda aşı noktasının altında daha yüksek oranlarda bulunması kombinasyonların polifenol durumları açısından uyuşur olduğunu göstermektedir. Gemlik, Sarı Haşebi, Halhalı (Hatay), Saurani and Kilis Yağlık olive varieties were grafted onto 1-year-old Gemlik rootstock. Samples were taken from the phloem tissues of grafted saplings that were 1 year old, dried in an oven at 65 °C, and made ready for analysis by grinding in a porcelain mortar. Then, the filtrate prepared by adding 10 ml of methanol to 1 g of sample was injected into HPLC and analyzed. The presence of phenolic compounds (protocatechic acid, caffeic acid, p-qumaric acid, ferulic acid, routine trihydrate and quercetin), which are abundant in olives, was determined. When the study results are examined; While protocatechic acid, caffeic acid, routine trihydrate and quercetin were detected at higher levels above the vaccination point, p-qumaric acid and ferulic acid were determined at higher levels below the vaccination point. The presence of phenolic compounds (ferulic acid and p-coumaric acid), which are considered to result in incompatibility in high amounts in the graft, are found in higher amounts below the graft point in all combinations, indicating that the combinations are compatible in terms of polyphenol status.Key Words: Incompatibility, floem, phenolic compounds, p-coumaric acid.
