Çalışmanın hayvan materyalini166 baş sağmal Holstein inek oluşturmuştur. İncelenen hayvanlar polimerazzincir reaksiyonu-restriksiyon parçacık uzunluk polimorfizmi (PCR-RFLP) yöntemiile yukarıda belirtilen genler yönünden genotiplendirilmiştir. İncelenen örneklerde FABP4 geni için G allel frekansının (%82), NR1H3 geni için G allel frekansının (%96) ve SCD geni için ise C allel frekansının (%73) en yüksek olduğugörülmüştür. Yapılan Hardy-Weinberg (HW) ki-kare analizleri sonucunda incelenenHolstein ineklerin FABP4 ve SCD genleri yönünden HW dengesindeoldukları, NR1H3 geni yönünden ise HWdengesinde olmadıkları görülmüştür. Çalışmasonunda, sütteki yağ oranı yönünden NR1H3genotipleri arasındaki farklılık (P<0.05), yağsız kuru madde oranı yönünden SCD genotipleri arasındaki farklılık (P<0.05)ve süt yoğunluğu yönünden FABP4genotipleri arasındaki farklılık (P<0.01)istatistiki olarak önemli bulunmuştur. Sonuçolarak incelenen genlerin süt kompozisyonu için yapılacak seleksiyonçalışmalarında kullanılabileceği düşünülmüştür. KW - Genotip frekansı KW - Holstein KW - polimorfizm KW - süt verim özellikleri N2 - The aim of this study was toinvestigate the relationship between genes that encode fattyacid-binding protein 4 (FABP4), nuclear receptor subfamily 1, group H, member3, (NR1H3) and stearoyl-CoA desaturase (SCD) proteins and some milk yieldtraits in Holstein cattle. A total of 166 Holstein dairy cows were used for thestudy. Genotyping was performed by polymerase chain reaction and restrictionlength polymorphism (PCR-RFLP) method. The frequencies of FABP4-G (82%), NR1H3-G(96%) and SCD-C (73%) alleles werefound to be high in the examined Holstein cows. According to chi-square testresults, the investigated Holstein cows were in Hardy-Weinberg (HW) equilibriumfor the FABP4 and SCD genes while significant deviationwas observed from HW equilibrium for the NR1H3gene. Regression analyses revealed that NR1H3genotypes were associated with milk fat (P<0.05), SCD genotypes were associated with dry matter without fat(P<0.05) and FABP4 genotypes wereassociated with milk density (P<0.01).Taken together, our results indicate that investigated SNPs may be used in theselection programs related with the milk composition traits in dairy cattle.
