Angiogenesisis the formation of new capillaries from existing blood vesselsthrough which oxygen and nutrients are transported to the tissuesto promote growth, and it plays a pivotal role in the invasionand metastasis of cancer caused by oncogenes. Even thoughthis process is useful in tissue repair and normal growth, it alsofacilitates the growth of cancer tissues and their metastasis viathe hematogenous route, and in this way, it speeds up metastasisand invasion. In the present study, we aimed to investigatethe relationship between angiogenesis and markers such asVEGF, MMP, TSP-1 and Endostatin in a CAPE-administeredgastric cancer cell in culture. Following CAPE administration,cytotoxicity was measured by MTT test. Angiogenic behavior ofcells was examined immunohistochemically by VEGF, MMP andEndostatin, and TSP – 1 level was measured by RT-PCR test. MTTtest showed that CAPE administered at a concentration of 0.5 μg/mL exerted a cytotoxic effect on a gastric cancer cell line. Geneexpression depicted by RT-PCR showed that VEGF, MMP and TSPlevels were decreased , while the endostatin level was increased.Changes in MMP, VEGF, TSP and endostatin suggest that CAPEhas a significant therapeutic effect on gastric cancer. In the lightof these findings, CAPE, which is a natural and inexpensivesubstance, could be effective in the treatment of cancer patientsand advanced studies need to be carried out. Buişlem doku onarımı ve normal büyüme için yararlı olsa da, kanserdokularının büyümesini ve hematojen yol yoluyla metastazınıkolaylaştırır ve bu şekilde metastazı ve invasyonu artırır. Buçalışmada mide kanseri hücre kültürüne uygulanan CAPE’ninVEGF, MMP, TSP-1 ve Endostatin gibi anjiyojenik ve antianjiyojenikbelirteçler arasındaki ilişkiyi araştırmayı amaçladık.CAPE uygulamasını takiben sitotoksisite, MTT testi ile ölçüldü.Hücrelerin anjiyojenik davranışı immünohistokimyasal olarakincelendi ve VEGF, MMP ve Endostatin ile TSP-1 seviyesi LCPCR testi ile ölçüldü. MTT testi, 0,5 μg/mL’lik bir konsantrasyondauygulanan CAPE’nin, bir mide kanseri hücre hattı üzerindesitotoksik bir etki yaptığını gösterdi. LC PCR ile gösterilengen ekspresyonu, VEGF, MMP ve TSP seviyelerinin azalırkenendostatin seviyesinin arttığını gösterdi. MMP, VEGF, TSP veendostatindeki değişiklikler, CAPE’nin mide kanseri üzerindeönemli bir terapötik etkiye sahip olduğunu düşündürmektedir. Bubulgular ışığında, doğal ve ucuz bir madde olan CAPE, kanserhastalarının tedavisinde etkili olabilir. Daha ileri çalışmalarınyapılması gerekir. CR - 1.
