Comparison of the Effects of Timing and Fixatives Type Used in the Detection of the Feline Ovary on the Protection of the Follicles Integrity

Abstract

study, we aimed to demonstrate the effect of 10% buffered formalin and Bouin solution on feline ovarian follicles and to determine the fixative
with the effect of the time elapsed between ligation and removal of tissue during OHE in feline ovaries.

Materials and Methods: Ovarian tissue samples from healthy, non-pregnant, owned domestic cats (aged 10-24 months) were obtained during
routine OHE. Half of each ovary was immersed into Bouin and 10% buffered formalin solution, followed by routine tissue processing after a oneday
fixation period, and stained with hematoxylin eosin. Healthy and degenerated primordial follicles were counted and compared between these
groups.

Results: Regardless of the fixative type, there was no difference in tissue preservation and integrity in the macroscopic and histological evaluation
of feline ovaries, which were removed during the operation with or without ligation and were directly fixed. Necrosis, congestion, or extravasation
were not observed in the ovaries. The follicles observed in the feline ovarian cortex fixed with Bouins’ solution were mostly healthy in appearance,
the majority of those fixed with formalin consisted of degenerated follicles.

Conclusion: In our study, it was revealed that short-term ligation did not affect the fixation of feline ovaries and that Bouin solution provided a
significantly better follicle and cellular integrity and preservation of the entire structure compared to formalin.

Kedi Ovaryumunun Tespitinde Zamanlamanın ve Kullanılan Fiksatiflerin Folikül Korunması Üzerindeki Etkilerinin Karşılaştırılması

Abstract

Amaç: Ovaryohisterektomi (OHE) ya da ovaryumun cerrahi operasyonlarında yapılan ligasyon doku bütünlüğü ve yapısını etkilemektedir. Özellikle
çıkarılan ovaryumlar kemoterapi/radyoterapi alan hastalarda dondurulup çözülerek tekrar nakil edilecek ise ligasyonun ovaryum üzerindeki etkisinin
histopatolojik analizi ve folikül görünüm ve yapılarının değerlendirilmesi önemli hale gelmektedir. Analizlerin doğru bir şekilde yapılabilmesi de
dokunun doğru solüsyonla fikse edilmiş olmasını gerektirir. Biz de bu nedenle çalışmamızda kedi ovaryumlarında OHE sırasında uygulanan ligasyon
sonrası dokunun uzaklaştırılması arasında geçen süre ile %10 tamponlu formalin ve Bouin fiksatiflerinin doku korunumu ve foliküller üzerine
etkilerinin ortaya konulması ve elde edilen bulgular doğrultusunda uygun fiksatifin belirlenmesini amaçladık.

Gereç ve Yöntem: Ankara Üniversitesi Veteriner Fakültesi, Doğum ve Jinekoloji Kliniği’ne rutin OHE amacıyla gelen sahipli kedilerden ovaryumların
alınması sırasında, tek taraflı ligasyon 3 dakika süreyle uygulandı. Her ovaryumun yarısı Bouin yarısı %10 tamponlu formalin solüsyonuna alınarak
bir günlük fiksasyon süresinden sonra rutin takibe alınıp hematoksilen eozinle boyandı ve sağlam ve dejenere primordiyal foliküller sayılarak
karşılaştırıldı.

Bulgular: Fiksatif tipinden bağımsız olarak operasyon sırasında ligasyon yapılarak ve yapılmaksızın çıkarılan ve doğrudan fiksatife alınan kedi
ovaryumlarının hem makroskobik açıdan hem de histolojik değerlendirilmesinde doku korunumu ve bütünlüğü bakımından incelendiğinde bir fark
ortaya çıkmadığı tespit edildi. Ovaryumlarda nekroz, konjesyon, ekstravazasyon izlenmedi. Fiksatiflerin doku korunumuna etkisi incelendiğinde;
Bouin ile fikse edilen kedi ovaryum korteksinde gözlenen foliküller çoğunlukla normal görünümde iken formalin ile fikse edilenlerin çoğunluğu
dejenere görünümde foliküllerden oluşmaktaydı.

Sonuç: Çalışmamızda kedi ovaryumlarının fiksasyonunda kısa süreli ligasyonun etkisinin bulunmadığı ve Bouin solüsyonunun formaline göre belirgin
olarak daha iyi bir folikül ve hücresel bütünlükle birlikte tüm yapının korunumunu sağladığı ortaya konulmuştur
