Antiproliferative Effects, Brdu Assay and Antioxidant Activities of Origanum sipyleum L. Extraction on Breast Cancer Cell Lines: Investigation in Vitro

Abstract

Origanum sipyleum L.   is commonly used for the treatment of cough, pain, cancer, and infectious diseases. In this study, the antioxidant and antiproliferative properties of this endemic species Origanum sipyleum L. (purple coral) were investigated. Quantitative analysis of the components of Origanum sipyleum L. was performed by HPLC. The antioxidant effects of Origanum sipyleum L.  ethanol extract was examined. Among the antioxidant effects, Total Polyphenolic Content Determination, Total Flavonoid Content Determination, Plasma Ferric Reducing Ability Test and Radical Scavenging Activity (DPPH) Determination were examined. The cytotoxic effect was determined using the XTT assay against MCF-7 (human breast adenocarcinoma cell line), MDA-MB-231 (human breast cancer cell line), and L929 (mouse  fibroblast cell line) cells. Calculated IC50 values of Origanium Sipyleum L. were 205±6.86 against MCF-7 cells for 24 h, 147±4.23 for 48 h, 197±4.15 for 24 h and 96± 3.46 against MDA-MB-231 cells, and 415±1.67 for 24 h 221±2.13 for 48 h, against L929, respectively. Antiproliferative properties of Origanium sipyleum L. was assessed by flow cytometry. MDA-MB-231 cell lines were used to determine cell proliferation and BrdU assay was analyzed spectrophotometrically. Results postulated that Origanum sipyleum L. had a high antioxidant and antiproliferative effects.
