Kanserde miRNA’ların Epigenetik Değişiklikleri ve Terapötik Uygulamaları

Abstract

Epigenetik, DNA dizisinde bir değişiklik olmaksızın gen ifadesindeki kalıtsal değişiklikler olarak tanımlanmaktadır. DNA metilasyonu gen
ifadesinin hücresel kontrolünde yer alırken, histon modifikasyonları da hücredeki transkripsiyonel aktivite ve kromatin yapıya ulaşabilirlik
kontrolünde fonksiyon göstermektedir. DNA metilasyonu ve histon modifikasyonları gibi epigenetik mekanizmaların ayrıcamiRNA’larınifade
sinidedüzenlediği belirlenmiştir. miRNA’lar hedef genlerinin ifadesini negatif yönde kontrol edebilenyaklaşık 22 nükleotid uzunluğunda küçük
RNA molekülleridir. miRNA ya da epigenetik mekanizmalarda ki tümör-ilişkili anomaliler insan kanserlerinde yaygın olarak saptanmaktadır.
Bu derlemede kanserde disregüle olan miRNA ve epigenetik mekanizmaların ilişkisi ve kanser terapisine yönelik miRNA’lar tanımlanmaya
çalışılmıştır.

Abstract

Epigenetics is identify as heritable changes in gene expression that do not appear a change in the DNA sequence. While DNA methylation involve in the control of cellular gene expression, histone modification function transcriptional activity in the cell and control the accessibility of chromatin structure. Epigenetic mechanisms such as DNA metylation and histone modifications also were determined that regulate the expressions of miRNAs. miRNAs are small RNA moleculesapproximately 22 nucleotide long that can negatively control their target genes expression.tumor-associated aberrations in the miRNA or epigenetic machineries are widely determined in human cancer. In this reviewhave been tried to define disregulated miRNA and epigenetic machineries relation between in cancer and miRNAs to cancer therapy
