COMPARISON OF IN VITRO REGENERATION POTENTIALS OF COWPEA (Vigna unguiculata (L.) Walp) AND SELLUKA (Vigna caracalla (L.) Verdc.) SPECIES

Abstract

Cowpea (Vigna unguiculata (L.) Walp), which belongs to the genus Vigna, has a very important role in both food and pharmaceutical uses in many countries of the world. Caracalla bean (V. caracalla (L.) Verdc.) whose flowers have volatile compounds is a perennial ornamental plant. The current study aimed to examine the differences between plant tissue culture stages and also regeneration potential in two different Vigna species, cowpea (Vigna unguiculata (L.) Walp) and caracalla bean (Vigna caracalla (L.) Verdc.). In order to determine the appropriate medium composition, callus cultures were established in MS and B5 media containing 2 mg/L 2,4-D with 0.2, 1 and 2 mg/L KIN concentrations. Then, the obtained calli were subcultured in media prepared by adding 10-50 g/L sucrose that gave the highest callus regeneration. The highest callus regeneration (76.66%) was obtained in MS medium containing 0.2 mg/L KIN in caracalla bean and 2 mg/L KIN in cowpea, while a higher regeneration rate was observed in MS nutrient medium than B5 for both species in all experiments. In addition, indirect root regeneration occurred in cowpea plant (46.67%), but a similar situation did not occur in caracalla bean. When the effect of sucrose was examined, the growth index tended to decrease as sucrose concentration increased in caracalla bean plants, whereas cowpea exhibited the opposite response. These results indicate that genotype plays a critical role in the in vitro culture response of Vigna species.
