Synthesis and biological evaluation of nicotinic acid derivatives as anti-hyperlipidemic potential in animal model
Abstract
Present research was designed to synthesize derivatives of nicotinic acid and to evaluate their effect against hyperlipidemic animal model. Derivatives of nicotinic acid were synthesized and characterized by UV, IR and 1 H-NMR techniques. Furthermore, these compounds were screened for anti-hyperlipidemic activity in male healthy rabbits divided into five groups. Each group containing five animals. Group I received only high cholesterol diet (150 days) and was named as untreated. While, group II-V received the high cholesterol diets from day 0 – 120 days followed by synthesized nicotinic acid derivatives, nicotinic acid and bezafibrate separately at dose (30 mg/day and 50 mg/day) intravenously. It was found that nicotinic acid derivatives receiving animals produced significant (p<0.01) anti- hyperlipidemic effect as compared to untreated group. The highest effect was shown by derivative II producing matchable results in lowering of all the lipid profile parameters. Hypolipidemic effect produced by derivative II was even better than nicotinic acid and bezafibrate. Furthermore, bezafibrate effect for lowering the low-density lipoprotein serum levels were found slight significant (p<0.05), while no significant p>0.05 in lowering total cholesterols and increasing HDL level. Present works established the significant hypolipidemic potential of newly synthesized nicotinic acid derivatives in male rabbits.
