Sonchus arvensis leaves water fraction alleviates inflammation symptoms in a rat model of acute gouty arthritis

Abstract

Gouty arthritis is inflammation of the joints that occurs in conditions of chronic hyperuricemia. When an acute attack of gout arthritis begins, oral colchicine is often given as the main treatment. However, colchicine can cause various side effects. Tempuyung (Sonchus arvensis) is a plant that is often found and easily cultivated in Indonesia, which has strong anti-inflammatory potential due to the presence of flavonoid compounds. This research aimed to investigate the anti-inflammatory activity of S. Arvensis leaves water fraction (SALWF) and detect flavonoids contained in it. An animal model of acute gouty arthritis was established using monosodium urate crystals injection. SALWF at 10 mg/kg BW was found to be effective in reducing the inflammation of the knee joint (p < 0.0001) and resulted in improvement in the H & E histological results. SALWF at 20 (p < 0.0001) and 40 mg/kg BW (p < 0.0005) also significantly alleviated swelling. Nevertheless, no histological improvements were observed at both doses. Eight flavonoids were successfully identified from LC-MS/MS, showing a significant proportion of baicalin and kaempferol 3-O-β-Dglucuronopyranoside. This research further establishes the use of SALWF for the management of acute gouty arthritis.
