Effects of Methionine and Cysteamine on Spermatological Parameters of Bee Spermatozoa Post Freezing-Thawing

Öz

In this study, it was aimed to determine the effects of methionine and cysteamine in the cryopreservation of drone semen (Apis mellifera). Semen samples were collected from sexually mature drones and pooled. The pooled semen was then divided into five equal volumes and diluted with diluents added at different concentrations of methionine (2.5/5 mMol), cysteamine (2.5/5 mMol) and control. Motility values post thaw; Met5, Cys2,5 and Cys5 groups had a possitive effect on compared to the the control group (P<0.05). It was observed that the antioxidant groups protected the plasma membrane integrity better than the control group (P<0.05). Acrosomal integrity rates was better in the methionine groups compared to the control group (P<0.05). Membrane lipid peroxidation was analyzed by measuring malondialdehyde (MDA) concentration after dissolution. It was observed that antioxidant-containing extenders had beneficial effects on honey bee semen parameters. Considering all semen parameters; The Met5 group was found to be the most effective group for freezing drone semen.

Arı Spermasının Dondurma-Çözdürme Sonrası Spermatolojik Parametreleri Üzerine Metiyonin ve Sisteaminin Etkileri

Öz

Bu çalışmada arı spermasının (Apis mellifera) dondurulmasında metiyonin ve sisteaminin etkilerinin belirlenmesi amaçlanmıştır. Sperma cinsel olarak olgun erkek arılardan toplandı ve pooling yapıldı. Pooling yapılan sperma beş eşit hacme bölündü ve farklı konsantrasyonlarda metiyonin (2.5 / 5 mMol) ve sisteamin (2.5 / 5 mMol) içeren ve içermeyen (kontrol) sulandırıcılar ile sulandırıldı. Eritme sonrası motilite değerleri; Met5, Sist2,5 ve Sist5 gruplarında kontrol grubuna göre daha üstün bulundu (P<0.05). Plazma membran bütünlüğünü antioksidan grupları kontrol grubuna göre daha iyi koruduğu görüldü (P<0.05). Akrozomal bütünlük oranları metiyonin gruplarında kontrol grubuna göre daha üstün bulundu (P<0.05). Membran lipid peroksidasyonu eritme sonrası malondialdehit (MDA) konsantrasyonu ölçülerek analiz edildi. Antioksidan içeren sulandırıcıların bal arısı sperm parametreleri üzerinde faydalı etkilere sahip olduğu görüldü. Tüm sperm parametreleri göz önüne alındığında; Met5 grubu arı spermasının dondurulmasında en etkili grup olduğu görüldü.
