                            <p>OBJECTIVE: In this study, it was aimed to evaluate the compatibility of white blood cell (WBC) count results obtained using an automatic blood count analyzer with the results obtained by manual microscopy, which is the reference method.MATERIAL AND METHODS: Between 2019 and 2020, sterile body fluid samples sent simultaneously to the medical biochemistry laboratory for automated body fluid cell count and to the medical microbiology laboratory for manual cell count were examined. During the manual microscopy stage, the samples were evaluated by looking at the microscope with a Thoma slide by two different researchers. WBC measurement was performed using the body fluid module (Sysmex Corporation, Kobe, Japan) on the Sysmex XN-10 blood counting analyzer.RESULTS: In our study, a total of 79 sterile body fluids (40 synovial fluid, 21 pleural fluid, 14 peritoneal fluid, 4 cerebrospinal fluid) were analyzed. &quot;Median (25%-75%) value was determined as 1520 (60-8340) cells/μL according to manual white blood cell count and 1615 (68-6091) cells/μL according to Sysmex XN-10. A statistically significant strong correlation was found between Sysmex XN-10 and manual microscopy. Statistical comparisons conducted with Passing-Bablok regression and Bland-Altman regression analyses revealed good agreement between manual white blood cell count and XN-10. Based on the 500 cells/μL cut-off value, the automated hematology analyzer demonstrated high diagnostic performance (sensitivity 93.9%, specificity 87.1%). The PPV, NPV, and overall accuracy were calculated as 92.0%, 90.0%, and 91.2%, respectively.CONCLUSIONS: Cell counting in sterile body fluids using an automatic analyzer is becoming increasingly common. In our study, it was determined that body samples gave generally compatible results with manual microscopy.</p></trans-abstract>
                                                                                                                                    <abstract><p>AMAÇ: Bu çalışmada otomatik kan sayım analizörü kullanılarak elde edilen beyaz küre hücre (WBC) sayım sonuçlarının referans yöntem olan manuel mikroskopi ile elde edilen sonuçlarla uyumunun değerlendirilmesi amaçlanmıştır.GEREÇ VE YÖNTEM: 2019-2020 yılları arasında tıbbi biyokimya laboratuvarına otomatize vücut sıvısı hücre sayımı, tıbbi mikrobiyoloji laboratuvarına manuel hücre sayımı yapılması amacıyla gönderilen steril vücut sıvısı örnekleri retrospektif olarak incelendi. Manuel mikroskopi aşamasında örnekler iki farklı araştırmacı tarafından Thoma lamı ile ışık mikroskopunda bakılarak değerlendirildi. Vücut sıvısının otomatize WBC ölçümü Sysmex XN-10 kan sayım cihazında vücut sıvısı modülü (Sysmex Corporation, Kobe, Japonya) kullanılarak analiz yapıldı.BULGULAR: Çalışmamızda toplam 79 steril vücut sıvısı (40 sinovyal sıvı, 21 plevra sıvı, 14 periton sıvısı, 4 beyin omurilik sıvısı) analiz edildi. Manuel beyaz küre hücre sayımına göre medyan (25%-75%) değeri 1520 (60-8340) hücre/μL, Sysmex XN-10&#039;a göre ise 1615 (68-6091) hücre/μL olarak saptandı. XN-10 ve manuel mikroskopi arasında istatistiksel olarak anlamlı güçlü bir korelasyon tespit edildi. Passing-Bablok regresyon ve Bland-Altman farklar analizi ile yapılan istatistiksel karşılaştırmalarda manuel beyaz küre hücre sayımı ve XN-10 arasında iyi bir uyum tespit edildi. 500 hücre/μL eşik değeriyle yapılan değerlendirmede, otomatik analizör yüksek tanısal performans göstermiştir (duyarlılık %93,9, özgüllük %87,1). PPV %92,0, NPV %90,0 ve doğruluk %91,2 olarak hesaplanmıştır.SONUÇ: Steril vücut sıvılarında hücre sayımının otomatik cihaz ile yapılması giderek yaygınlaşmaktadır. Yaptığımız çalışmada vücut örneklerinde manuel mikroskopiyle genel olarak uyumlu sonuçlar verdiği saptandı.</p></abstract>
