Koç Spermasının Dondurulmasında Oksidatif Stres Üzerine Trolox ve Taurinin Etkisi

Abstract

Bu çalışmada Pırlak koç spermasının üreme mevsimi dışında dondurulmasında sperma sulandırıcısına eklenen antioksidan özelliğe sahip trolox ve taurinin çözüm sonu spermatolojik parametreler üzerine etkisinin ortaya konması amaçlandı. Bu amaçla 10 baş Pırlak koçtan haftada iki kez olacak şekilde suni vagen yardımı ile spermalar toplandı. Normospermik özellik gösteren numuneler birleştirilerek sırası ile kontrol, 1 mM Trolox ve 50 mM Taurin grupları oluşturuldu. Gruplar +40 C’de 2 saat ekilibrasyon işlemi sonrası, 0.25 ml’lik payetlere çekildikten sonra -1200 C sıvı azot buharında dondurularak, sıvı azot içerisinde saklandı.Çözüm sonu spermatolojik değerlendirmelerden motilite sübjektif yöntem ile, plazma membran bütünlüğü (PMAI), yüksek mitokondriyal aktivite düzeyi (HMMP) ve oksidatif hasar düzeyi (MITOSOX+) flow sitometrik yöntem ile incelendi. Çözüm sonu motilite oranı 1 mM Trolox (%50,00±3,89)ve 50 mM Taurin (%60,62±2,57) gruplarında kontrol (35,00±2,11) grubuna yüksek elde edildi (p<0.05). Taurin 50 mM (%32,42±0,72) çözüm sonu en yüksek PMAI oranı ile kontrol grubuna kıyasla istatistiksel olarak önemli bulundu (P<0.05). Antioksidan ilave edilen gruplarda HMMP değerleri kontrol grubuna göre yüksek bulunurken, oksidatif hasar düzeyi en düşük grup Taurin 50 mM olarak belirlendi (P<0.05). Sonuç olarak Pırlak koç spermasının sezon dışı dondurulmasında sulandırıcıya ilave edilen Trolox ve Taurinin çözüm sonu motilite, HMMP parametreleri bakımından olumlu etkileri olabileceği kanısına varılırken 50 mM Taurin ilavesinin spermanın dondurulması sonucu oluşan oksidatif stres üzerine koruyucu etkinliğinin oluştuğu belirlendi.

Effects of Trolox and Taurin on Oxidative Stress During Ram Semen Cryopreservation

Abstract

In this study, ıt was aimed the investigate the effects on Pirlak ram spermatological parameters added sperm extender which antioxidant properties of Trolox and Taurin. For this aim semen was collected with the aid of artificial vagina twice a week from 10 Pirlak ram. Semen which were normospermic pooled and occurred three group; control, 1 mM Trolox, 50 mM Taurin. Groups were equilibrated about 2 hours at + 4 ° C, frozen in liquid nitrogen vapor (-120 ° C), stored in liquid nitrogen. Pos-thawed spermatological examinations; motility was recorded subjectively, plasma membrane integrity (PMAI), high mitochondrial membrane potential (HMMP) and mitochondirial oxidative damage level (MITOSOX+) were evaluated by flow cytometer. Post-thawed motility ratio was higher in 1 mM Trolox (%50.00±3.89) and 50 mM Taurin groups compared the control (35.00±2.11) group (p<0.05). Taurin 50 mM (%32.42±0.72) was highest PMAI ratio and it was significantly important with control group (P<0.05). HMMP levels were higher in antioxidant groups when compared the control group and also lowest mitochondirial oxidative damage level was seen in the 50 mM Taurin (P<0.05). Finally, when freezing of Pirlak ram semen in the out of breeding season Trolox and Taurin which were added the semen extender was ameliorating effect on post thaw motility and HMMP levels. Behind this Taurin 50 mM gave the best preventive effect on oxidative damage.
