The effects of lower and higher dosage cisaprid, bethanechol and theirs  combinations on abomasal motility and emptying rate of healthy calves

Öz

Sığırlarda paralitik ileus, sekum dilatasyonu/dislokasyonu ve abomazum hipotonisi gibi motilite yetersizliğine sebep olan hastalıklar önemli verim kayıplarına neden olabilmektedir. Sığırlarda gastrointestinal motilite düzenleyicileri ile ilgili bilgiler yetersizdir. Sunulan bu araştırmada sisaprid ve betanekol’un iki farklı dozunun ve kombinasyonların sağlıklı buzağıların abomazum motilitesi ve boşalma oranı üzerindeki etkinlikleri dört farklı metotla belirlendi. Araştırmada hayvan materyali olarak doğumu takiben 4-5 günlük, sağlıklı toplam 10 adet buzağı kullanıldı. Her buzağıda yüksek ve düşük doz sisaprid, betanekol ve kombinasyonlarının uygulanmasını takiben 30 dakika sonra süt ikamesi içirildi. Buzağılardan 12 saat süresince aralıklı olarak kan örnekleri alındı. Abomazal motilite ve boşalma oranı asetaminofen absorbsiyon test, ultrasonografik metot, abomazal intraluminal basınç ve glikoz değişim grafiği ile değerlendirildi. Sağlıklı toplam 5 adet (4-5 günlük) buzağı kullanıldı. Buzağılar süt ikamesi (60 ml/kg) ile günlük olarak aynı saatlerde beslendi. Buzağılardan deneme süresince aralıklı olarak kan örnekleri alındı. Abomazal motilite ve boşalma oranı asetaminofen absorbsiyon test, ultrasonografik metot, abomazal intraluminal basınç ve glikoz değişim grafiği ile değerlendirildi. Ultrasonoğrafik yarı ömür ve β sonuçları irdelendiğinde, sadece süt ikamesi verilen kontrol grubuna göre tüm ilaç uygulamalarının bu değerlerinin daha küçük olduğu (p<0.05) belirlendi. Asetominofen zaman-konsantrasyon eğrileri incelendiğinde ise, ilaçların ortalama 120-300. dakikalar arasında en yüksek yoğunluğa ulaştığı belirlendi. Abomazal boşalma hız ve derecesinin göstergesi olarak belirlenen AUC 0-120 değerinin ise sisaprid ve kontrol gruplarına kıyasla, betanekolün tek başına ve kombine uygulandığı gruplarda önemli olduğu(p<0.05) tespit edildi. Sonuç olarak; ultrasonografik yarı ömür ve asetominofene absorbsiyon testin değerlendirilmesi doğrultusunda betanekol uygulaması sonrasında elde edilen AUC 0-120 değerlerinin sisaprid ve kombine uygulamalara göre daha hızlı abomazal boşalmaya sebebiyet verdiği tespit edilmiştir.

Sağlıklı buzağılarda yüksek ve düşük doz sisaprid ve betanekol’ün abomazum motilite ve boşalma oranına etkisi

Sığırlarda paralitik ileus, sekum dilatasyonu/dislokasyonu ve abomazum hipotonisi gibi hastalıklar önemli  oranda  verim  kayıplarına  neden  olabilmektedir.  Ruminantlarda  gastrointestinal  motilite düzenleyicileri hakkında bilgiler yetersizdir. Bu araştırmada sisaprid ve betanekol’un iki farklı dozunun ve kombinasyonların sağlıklı buzağıların abomazum motilitesi ve boşalma oranı üzerindeki etkinlikleri dört farklı  metotla belirlendi.  Hayvan  materyalini  doğumu  takiben 4‐5  günlük  yaşta,  sağlıklı  toplam 10  adet buzağı  oluşturdu.  Her  buzağıda  yüksek  ve  düşük  doz  sisaprid,  betanekol  ve  kombinasyonlarının uygulanmasını takiben 30 dakika sonra süt ikamesi içirildi. Buzağılardan 12 saat süresince aralıklı olarak kan örnekleri alındı. Abomazal motilite ve boşalma oranı asetaminofen absorbsiyon testi, ultrasonografik metot, abomazal intraluminal basınç ve glikoz değişim grafiği ile değerlendirildi. Kontrol amacıyla sağlıklı toplam  5  adet  (4‐5  günlük)  buzağı  kullanıldı.  Buzağılar  süt  ikamesi  (60  ml/kg)  ile  günlük  olarak  aynı saatlerde beslendi. Ultrasonoğrafik yarı ömür ve β sonuçları irdelendiğinde, sadece süt ikamesi verilen kontrol  grubuna  göre  tüm  ilaç  uygulamalarının  bu  değerlerinin  daha  küçük  olduğu  (p<0.05)  belirlendi. Asetominofen zaman‐konsantrasyon eğrilerinde, ilaçların ortalama 120‐300. dakikalar arasında en yüksek yoğunluğa  ulaştığı  belirlendi.  Abomazal  boşalma  hız  ve  derecesinin  belirlenmesinde  Area  Under  Curve (AUC, biyoyararlanım eğrisi altında kalan alan) değerlendirildi ve belirlenen AUC 0‐120 değerinin sisaprid ve kontrol gruplarına kıyasla, betanekolün tek başına ve kombine uygulandığı gruplarda istatistiksel açıdan önemli olduğu (p<0.05) tespit edildi. Sonuç olarak; ultrasonografik yarı ömür ve asetominofen absorbsiyon testinin değerlendirilmesi doğrultusunda her iki dozda betanekol uygulaması sonrasında elde edilen AUC 0‐120 değerlerinin sisaprid ve kombine uygulamalara göre daha hızlı abomazal boşalmaya sebebiyet verdiği tespit edilmiştir. Anahtar kelimeler: buzağı, abomazal boşalma, sisaprid, batanekol, asetaminofen absorbsiyon testi
