Evaluation of the Effects of Asprosin on Cell Viability and DNA Damage on Human Glioblastoma and Mouse Neuroblastoma Cell Lines

Abstract

Asprosin is a peptide hormone secreted from white adipose tissue during fasting and plays a role in the regulation of energy metabolism. Its ability to cross the blood–brain barrier and exert effects on the central nervous system has attracted attention regarding neurological disorders and brain tumors. Recent studies suggest that asprosin may also be involved in tumor biology. This study investigated the effects of asprosin on cell viability and DNA damage in human glioblastoma (U87-MG) and mouse neuroblastoma (N2a) cell lines. Cells were treated with 1, 10, and 100 ng/mL asprosin. Cell viability was assessed using the 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) assay, while DNA damage was evaluated by the Comet assay. Data were analyzed using GraphPad Prism 9. One-way analysis of variance (ANOVA) followed by Tukey's post hoc test was used for statistical analysis, and p<0.05 was considered statistically significant. Asprosin reduced cell viability in both cell lines. A significant decrease was observed at 100 ng/mL in U87-MG cells and at 10 and 100 ng/mL in N2a cells. Comet assay results demonstrated that asprosin significantly increased DNA damage in both cell lines. In conclusion, asprosin exhibited cytotoxic and genotoxic effects in tumor cells. These findings suggest that asprosin may represent a potential therapeutic candidate for the treatment of glioblastoma and neuroblastoma.
