                            <p>ObjectiveWhile chemotherapeutic drugs are effective at eliminatingcancer cells, they can also damage healthycells. The aim of this study was to analyze the phenoliccomponent content of wheatgrass extract (Triticumaestivum L.) and to determine the effect of thisextract against DNA damage caused by cisplatin andetoposide used in chemotherapeutic treatment inhealthy cells will.Material and MethodSprouted wheat methanol extract was prepared andanalysis of phenolic components was performed byHPLC (high performance liquid chromatography).IC50 (half maximal inhibitory concentration) and LD50(median lethal dose) values were calculated by applyinga cell viability assay based on wheat extractconcentration. Cells were incubated at these determinedconcentration levels and the presence of DNAdamage was assessed by the Comet method.ResultsAs a result of the phenol component analysis, p-hydroxybenzoicacid was determined in the highestamount and o-coumaric acid in the lowest amount.The values for wheatgrass extract, etoposide andcisplatin at different concentrations applied to lymphocytecells were found to be IC50=204.6 μg/mL,LD50=15.84 μg/mL and LD50=24.51 μg/mL, respectively.As a result of comet analysis, it was foundthat etoposide LD50 and etoposide LD50+wheatgrassextract IC50 group were statistically significant(p0.05). Similar to this result, cisplatin LD50 andcisplatin LD50+wheatgrass extract IC50 group werefound to be statistically significant compared to thecontrol group (p 0.05).ConclusionAlthough wheatgrass was observed in our study tocause a reduction in DNA damage caused by etoposideand cisplatin, no statistical significance wasfound.</p></trans-abstract>
                                                                                                                                    <abstract><p>AmaçKemoterapötik ilaçlar kanser hücrelerinin ortadankaldırılmasında etkili iken aynı zamanda sağlıklı hücrelerdede hasar oluşturabilmektedir. Bu çalışmada,buğday çimi (Triticum aestivum L.) ekstraktının fenolikbileşen içeriğinin analizi ve bu ekstraktın kemoterapötiktedavide kullanılan sisplatin ve etoposid’in sağlıklıhücrelerde oluşturduğu DNA hasarına karşı etkisininbelirlenmesi amaçlanmıştır.Gereç ve YöntemÇimlendirilmiş buğdayların metanol ekstraktı hazırlanarakHPLC (yüksek performanslı sıvı kromatografisi)ile fenolik bileşen analizi yapıldı. Buğday ekstraktıkonsantrasyonuna bağlı hücre canlılık testi uygulanarakIC50 (Yarı maksimum inhibitör konsantrasyonu) veLD50 (ortalama öldürücü doz) değerleri hesaplandı.Belirlenen bu konsantrasyon değerleri ile hücreler inkübeedilerek DNA hasarı varlığı Comet metodu iledeğerlendirildi.BulgularFenolik bileşen analizi sonucunda p-hidroksibenzoikasit en yüksek miktarda, o-kumarik asit ise en düşükdüzeyde tespit edildi. Lenfosit hücrelerine uygulananfarklı konsantrasyonlardaki buğday çimi ekstraktı,etoposid ve sisplatin için değerler sırasıyla IC50=204,6μg/mL, LD50=15,84 μg/mL ve LD50=24,51 μg/mL olarakbulundu. Comet analizi sonucunda kontrol grubunakıyasla, etoposid LD50 ve etoposid LD50+buğdayçimi ekstraktı IC50 grubu istatistiksel olarak anlamlıbulunurken (p0,05). Busonuca benzer olarak kontrol grubuna kıyasla, sisplatinLD50 ve sisplatin LD50+buğday çimi ekstraktı IC50grubu istatistiksel olarak anlamlı bulunurken (p0,05).SonuçÇalışmamızda buğday çiminin etoposid ve sisplatinnedeni ile oluşan DNA hasarında azalmaya neden olduğugörülmüş olmasına rağmen istatistiksel olarakanlamlılık saptanmamıştır.</p></abstract>
