Türkiye’de Kültürü Yapılan İskenderiye Üçgülü (Trifolium alexandrinum L.) ve Iran Üçgülü (T. resupinatum L.) Türlerinin Karyotip Özellikleri

Abstract

Türkiye’de kültürü yapılan ve birbirinden farklı ekolojik kosullara adaptasyon gösteren İskenderiye üçgülü (Trifolium alexandrinum L.)
ve İran üçgülü (Trifolium resupinatum L) türlerinin karyotip analizi ve kromozom incelemeleri A. Ü. Ziraat Fakültesi Tarla Bitkileri Bölümü
Sitoloji Laboratuvarı’nda yürütülmüştür. Çimlenen tohumlardan elde edilen kök ucları %1 ά-bromonaftalin ve 8- hidroxykuinolin
çözeltilerinde ilk işleme tabi tutularak, Sonra aseto demir hematoksilin ile boyanmıştır. Elde edilen sonuçlara göre; her iki tür için de önceki
calışmalara uygun olarak kromozom sayıları 2n = 2x= 16 olarak doğrulanmıştır. İskenderiye üçgülü (Trifolium alexandrinum L.) ve İran
üçgülü (Trifolium resupinatum L) ortalama kromozom boyları sırasıyla 3.45 ± 0.06 ve 4.19 ± 0.13 olarak bulunmuştur. Önceki calışmalarda
tam olarak ortaya konulamadığı bildirilen İskenderiye üçgülü kromozomlarında satelit olgusu iki kromozom çiftinde tespit edilmiş, 1
numaralı kromozom 3.26 ± 0.37 kol oranı ile submetasentrik, 7 numaralı kromozom ise 1.23 ± 0.10 kol oranı ile metasentrik tipte kromozom
olarak bulunmuştur. Satelit uzunlukları sırasıyla 1.51 ve 0.95 µm’dir. İran üçgülü. kromozomlarında ise ilk kez bir kromozom çiftinde
heteromorfizme rastlanmıştır. Bu olgu karyotipte 2 numaralı kromozomun homologları olarak 2(1) numaralı olan 1.16 ± 0.10 kol oranı ile
metasentrik iken, 2(2) numaralı homolog ise 2.17 ± 0.52 submetasentrik olarak belirlenmiştir.

Karyotype Features of Berseem Clover (T. alexandrinum L.) and Persian Clover (T. resupinatum L.) Species Which Cultivated in Turkey

Abstract

Karyomorphological studies were carried out on two putative and cultivated Trifolium species in Turkey; berseem (Trifolium alexandrinum L.) and persian clover (T. resupinatum L.), these two species extensively for their adaptation to different ecological conditions are cultivated as forage plant in Turkey. Meristematic cells of root tips (1 cm in length) were obtained from germinated seeds in petri dishes and pretreated in 1% ά- Bromonaphthalene and 8-Hydroxyquinoline solutions and stained in aceto iron hematoxyline with macerating stained roots by means of the enzyme cytase. The results showed that the Somatic chromosomes of these species are contains equal diploid chromosome number 2n=2x= 16; Avarage chromosome length were 3.45± 0.06 and 4.19± 0.13 for berseem and persian clover, respectively. A phenomenon having satelite were found for four chromosome of berseem clover which was reported undefinite in previous studies; chromosome 1 and chromosome 7 were found submetacentric with the arm ratio 3.26± 0.37 and metacentric with the arm ratio 1.23± 0.10, respectively. Sattelite lentghs of these chromosomes are 1.51 and 0.95 µm. In persian clover chromosomes one pair was found having heteromorphism. This phenomenon was determined in karyotype that as the homologous of chromosome 2, 2(1) was metacentric with the arm ratio 1.16± 0.10 and 2(2) was submetacentric with the arm ratio 2.17± 0.52
