Localization of 1 integrin and fibronectin on spermatogenic cells in rat testis

Öz

Aim: Fibronectin interacts with its receptor, 1
integrin, and takes place in cell adhesion,
migration and proliferation. In this study, we
examined the distribution of fibronectin and 1
integrin of testis tissues in different
developmental stages with
immunohistochemical methods.
Material and methods: Wistar rats were used
in the study and divided into 8 groups
according to their ages: newborn, 3 days, 10
days, 15 days, 21 days, 1 month, 2 months and
adult. Testis tissues were examined
immunohistochemically for fibronectin and 1
integrin.
Results: The best fibronectin reactions
occurred in peritubular area, basement
membrane and myoid cell layer and these
reactions became more apparent in later stages.
Besides we observed that there was no reaction
in seminiferous tubule epithelium till adult
stage. In adult rat testis tissue, fibronectin
immune staining was mild to weak in cell
layers of seminiferous epithelium. 1 integrin
immune staining was diffusely strong in
peritubular tissue and spermatogonia from
seminiferous epithelium. Reaction was
restricted to cytoplasm and membranes of the
germinal cells.
Conclusion: As a result, fibronectin in testis is
more functional in cell-basement membrane
and basament membrane-interstitial tissue
interaction than cell-cell interaction; and 1
integrin is essential in cell-cell interaction and
spermatogenesis.

Testiste Spermatogenik Hücrelerde 1 İntegrin ve Fibronektin Dağılımı

Öz

Amaç: Fibronektin β1 integrinle etkileşerek
hücre adezyonu, göçü ve proliferasyonunda
görev alır. Bu çalışmada gelişmekte olan testis
dokusunda 1 integrin ve fibronektin
dağılımını immünohistokimyasal yöntemle
göstermek amaçlandı.
Gereç ve yöntem: Çalışmada; Wistar cinsi
erkek sıçanlar kullanıldı. Sıçanlar, yaşlarına
bağlı olarak ver her grupta 7 adet olmak üzere
yenidoğan, 3, 10, 15, 21 günlük, 1, 2 aylık ve
ergin olmak üzere 8 gruba ayrıldı. Dokulara
fibronektin ve 1 integrin için
immünhistokimyasal yöntem uygulandı.
Bulgular: Testiste fibronektinin bir bazal
membran ve bağ doku bileşeni olarak
seminifer tüplerin bütünlüğüne katkıda
bulunduğu ve hücre-hücre etkileşiminden çok
hücre-bazal membran ve bazal membraninterstisyel doku etkileşiminde işlevsel olduğu
görüldü. Fibronektin tutulumunun en belirgin
izlendiği alanların peritübüler alan, bazal
membran ve myoid hücre katmanı olduğu
görüldü. 1 integrin tutulumunun, özellikle
gelişimin ileri evrelerinde, peritübüler dokuda
ve bazı spermatogenik seri hücrelerinde
belirgin olduğu izlendi.
Sonuç: β1 integrinin ise daha çok hücre-hücre
etkileşiminde ve dolayısıyla spermatogeneziste
etkin olabileceği kanısına varıldı.
