Investigation of Protective Effect of Hypericum Perforatum on Ethanol-induced Acute Gastric Mucosal Injury in Rats

Öz

Objective: This study was conducted
to evaluate the antiapoptototic and antiinflammatory effect of Hypericum
perforatum in the rats induced gastric ulcer by absolute ethanol. Materials and
Methods: Forty male Wistar albino rats were used in this study. Rats were
divided four group, randomly. Group I (Control, n=10), Group II (Ethanol, n=10,
1 mL ethanol, orally, 90 min)  Group III
(H. perforatum + ethanol, 1 mL H. perforatum, 120 min; 1 mL ethanol, 90
min, orally), Group IV (Olive oil + ethanol, 1 mL olive oil, 120 min; 1 mL
ethanol, 90 min, orally). Gastric ulcer score 
was determined in gastric mucosa by morphometrically. Histopatological
staining, Immunohistochemical iNOS and PCNA staining, apoptotic TUNEL staining,
gastric IL-10 and IL-6 expression (ELISA) and SOD, MDA, CAT levels were
assessed in gastric tissue. Results: The results showed that ethanol incuded
gastric damage, higher gastric ulcer index score, increased proinflammatory
IL-6 level, elevated number of iNOS and TUNEL-positive stained cell number as
well as higher MDA level in the group II (ethanol). Pre-treatment of H. perforatum and olive oil (group III
and IV) significantly attenuated the gastric lesions as compared to the group
II (ethanol). Conclusion: It was concluded that H. perforatum and olive oil may represents a potential therapeutic
option to reduce the risk of gastric mucosal ulceration.

Etanol Uyarımlı Sıçan Akut Mide Mukoza Hasar Modelinde Hypericum Perforatum'un Koruyucu Etkilerinin İncelenmesi

Öz

Amaç:
Bu çalışma sıçanlarda absolü alkol ile oluşturulan gastrik ülserde H. perforatum'un antiapoptotik ve
antiinflamatuvar etkilerini değerlendirmek amacıyla yapılmıştır. Gereç ve
Yöntem: Bu çalışmada 40 adet erkek Wistar albino sıçan kullanılmıştır. Sıçanlar
rastgele 4 gruba ayrılmıştır. Grup I (Kontrol grubu, n=10), Grup II (Etanol
grubu, n=10, 1 mL etanol, oral yolla, 90 dk), Grup III (H. perforatum+Etanol grubu, n=10, 1 mL H.perforatum, 120 dk; 1 mL etanol, 120 dk, oral yolla), Grup IV
(Zeytinyağı+Etanol, 1 mL zeytinyağı, 120 dk; 1 mL etanol, 120 dk, oral yolla).
Gastrik ülser skoru mide mukozasında ülserasyon alanlarının ölçülmesiyle
hesaplandı. Gastrik dokuda histopatolojik boyamalar, immunohistokimyasal İNOS
ve PCNA boyamaları, TUNEL boyaması, IL-6 ve IL-10 ifadeleri (ELISA) ve MDA, SOD
ve CAT enzim seviyeleri değerlendirildi. Bulgular:Sonuçlara incelendiğinde,
etanol uyarımlı modelde gastrik hasar; yüksek ülser indeksi skoru, yükselmiş
proinflamatuvar IL-6 seviyesi, fazla sayıda TUNEL-pozitif hücre, artmış iNOS
ifadesi ve yüksek MDA seviyesi bulguları tespit edildi. H. perforatum ve Zeytinyağı ile ön tedavi yapılmış gruplarda
gastrik lezyonların önemli derecede azaldığı tespit edildi. Sonuç: Sonuç olarak
H. perforatum ve zeytinyağı gastrik
mukozal ülserasyon riskini azaltmak için potansiyel bir tedavi seçeneği
olabilir.
