OLAY YERİNDE BAKTERİYEL KONTAMİNASYONA MARUZ KALAN DIŞKI VE İDRAR ÖRNEKLERİNDEN KİMLİKLENDİRME

Abstract

Amaç: Adli vakalarda kan, semen ve tükürük gibi biyolojik materyallerin kullanımı oldukça yaygındır. Diğer önemli biyolojik örneklerden olan idrar ve dışkı numuneleri, adli amaçlı karışık örneklerin incelenmesinde, doping kontrolünde, uyuşturucu tarama programlarında, suç yerinin tespitinde, ölüm sebebinin anlaşılmasında, cinsel saldırı veya taciz davalarında kullanılmaktadır. Delil olan dışkı numunesi, en yaygın olarak fiili livata olgularından veya anal yolla girmiş herhangi bir materyalden alınacak örnekten elde edilebilir. Yöntemler: Çalışmada, 30 gönüllü kişiden rıza formu doldurularak temin edilen dışkı ve idrar örnekleri ile çalışıldı. Gönüllülerden alınan materyallerin yanında, kontrol grubu için ağız içinden sürüntü örnekleri alındı. Kontrol DNA örneklerinden Chelex ile DNA ekstraksiyonu, çeşitli ticari kitlerle miktar tayini ve amplifikasyonu yapılıp elektroforezle tiplenmesi gerçekleştirildi. DNA toplama işlemi, çeşitli şartlar altında tutulmuş idrar ve dışkı örneklerinin yüzeyinden steril nemli koton swapla yapıldı. Dışkıdan elde edilen DNA’ların ekstraksiyonu ticari kit ile, idrardan elde edilenlerin ki ise Chelex yöntemi ile yapıldı. Dışkı ve idrardan elde edilen DNA için miktar tayini ve amplifikasyonu çeşitli ticari kitler ile yapılıp, kapiler elektroforez ile tiplendirildi. Bulgular: Profillemesi yapılan idrar örneklerinde, kadınlara ait örneklerin erkeklere ait örneklerden daha iyi sonuçlar verdiği gözlenmiştir. Kontaminasyonun kimliklendirme üzerine olan etkisi incelendiğinde, bakterilerin biyolojik materyallere salgıladıkları enzimlerle zarar verdiği, ancak yürütme aşamasında, kullanılan kitlerdeki lokusların insana özgü olmasından dolayı çalışmayı etkilemediği gözlenmiştir. Sonuç: İdrar ve dışkı örneklerinin, diğer biyolojik örnekler gibi kimliklendirmede başarılı sonuçlar verdiği görülmüştür.

IDENTIFICATION FROM CONTAMINATED FECAL AND URINE SAMPLES AT THE CRIME SCENE

Abstract

Objective: Biological materials such as blood, semen, and saliva are quite common in criminal cases. The other biological samples as urine and gaita are used in examination of mixed samples for forensic purposes, doping control, drug screening programmes, crime scene investigation CSI , finding out cause of death and sexual abuses/assaults. Mostly gaita is obtained from cases in the act of buggery or anal assaults. Methods: In the present study, 30 volunteers have given their urine and gaita samples after getting their informed consents. For control group buccal swaps were studied. In controls; DNA extraction was analysed with Chelex, besides quantitation and amplification were analysed with some commercial kits; following these typing was analysed with electrophoresis. DNA from surfaces of urine and gaita samples which were kept in some conditions was obtained by sterile wet coton swaps. DNA extraction for gaita with commercial kit and Chelex for urine were analysed. For DNA obtained from urine and gaita, quantitation and amplification were analysed with some commercial kits and typing was analysed with capiller electrophoresis. Results: It is observed that urine samples of women showed out better results than men. When effect of contamination on identification was examined, bacteria secreting enzymes damaged biological materials, but loci in kits used in step of profiling, being specific to human, did not effect the study. Conclusion: Urine and gaita could present succesful results such as the other biolgical materials.
