Cobas - Bio® Santrifügal Analizleme Gereci ile İdrarda Tetrahydrocannabinol Türevlerinin Belirtilmesi

Abstract

İdrarda cannabinoid türevlerinin aranması için kullanılan EMIT® Cannabinoid Urine Assay kitinin tüm deney basamakları Cobas-Bio® santrifügal analizleme gerecine programlandı. Deneylerde kullanılması gereken antikor-substrat ve enzim derişimleri belirlendi. Standart eğrinin tekrarlanabilirliği ve doğrusal olduğu bölüm incelendi. 20.0, 30.0 ve 50.0 ng/mL derişimlerinde 11-nor-A8-THC-9-karboksilli asid (THCCOOH) içeren idrar örneklerine belirli miktarlarda THC-COOH katılarak deneyin gerielde analizi yapıldı. Elde edilen bulgulara dayanarak, duyarhlık alt sıırıın 20.0 ng/mL THCCOOH olduğu, 100 ng/mL'den fazla THC-COOH içeren idrarların seyretildikten sonra yeniden çalışılması gerektiği, yanlış pozitif sonuç alınmadığı ve deneyin, kısa zaman içerisinde fazla sayıda idrarda tarama (screening) amacıyla kullanılmaya elverişli olduğu sonucuna varıldı.
