An Experimental Study on Pathogenesis of Infectious Pancreatic Necrosis in Rainbout Trout: The Role of Tip 1 Interferones in Innate Immunity

Abstract

Backround/Aim: Infectious Pancreatic Necrosis IPN is an acute, contagious, systemic viral disease characterized by necrosis of the pancreas in different fish species. The agent is the IPN virus IPNV in the Aquabirnavirus family from the RNA group. Material and Method: An experimental infection with IPNV isolate isolated from a trout company in Turkey was carried out. After necropsy applications, the tissues were embedded in paraffin and prepared sections 5 µm in diameter. For determination of IPNV antigen and expression of tip-1 interferon and their receptor, enzyme based immunohistochemistry was applied on tissue sections. Conclusion: Experimental infection of trout with IPNV showed that although expression of Tip-1 interferon and its receptors in the infected tissues was not clearly visible, it partially blocked viral replication and could not prevent the establishment of persistent infection.

Gökkuşağı Alabalıklarında İnfeksiyöz Pankreatik Nekroz’un Patogenezisi Üzerine Deneysel Bir Araştırma: Tip 1 İnterferonların İnnate Doğal Bağışıklıktaki Rolü

Abstract

Özbilgi/Amaç: Enfeksiyöz Pankreatik Nekroz IPN , değişik balık türlerinde pankreasın nekrozu ile karakterize, akut, bulaşıcı, sistemik viral bir hastalıktır. Etken, RNA grubundan Aquabirnavirüs ailesinde IPN virusu IPNV ’dur. Bu çalışma ile, Türkiye’deki bir alabalık işletmesinden izole edilen IPNV izolatı ile deneysel enfeksiyon gerçekleştirilerek; alabalıklarda, doğal bağışıklıkta önemli yeri olan virus-konakçı hücre ilişkileri interaksiyonları ’nde interferonların rolü araştırılmıştır. Materyal ve Metot: Deneysel enfeksiyonu takiben nekropsileri gerçekleştirilen hayvanlarda dokular parafinde bloklandı. Viral antijen ve tip1 interferon ve reseptörlerinin lokalizasyonu ve yoğunluğu için enzim tabanlı immunohistokimyasal boyamalar gerçekleştirildi. Sonuç: Alabalıkların IPNV ile deneysel enfeksiyonunda, Tip-1 interferon ve reseptörlerinin enfekte dokularda ekpresyonları belirgin şekilde görülmesine karşın, viral replikasyonu kısmen bloke ettiği ve persiste enfeksiyonun kurulumunun önüne geçemediği görülmüştür.
