Methylenetetrahydrofolate Reductase Activity and Folate Metabolism

Abstract

Folate is a vital B vitamin which is easily water-soluble. It is a natural source which is found in the herbal and animal foods. Folate has important duties in the human metabolism, one of them is the adjustment of the level of plasma homocysteine. Reduction in MTHFR (methylenetetrahydrofolate reductase),which is in charge of the metabolism of homocysteine activity affects the level of homocysteine. Therefore MTHFR is an important enzyme in folate metabolism. Some of the mutations occurring in the MTHFR gene is a risk factor for various diseases and may be caused the hyperhomocysteinemia or the homocystinuria, and they also may lead to metabolic problems. MTHFR is effective in the important pathways such as DNA synthesis, methylation reactions and synthesis of RNA. C677T and A1298C are the most commonly occurring polymorphisms in the gene of MTHFR. The frequency of these polymorphisms show differences in the populations. MTHFR, folate distribution, metabolism of homocysteine and S-adenosylmethionine, by the MTHFR methylation the genetic defects have the potential of affecting the risk of disease in the negative or positive way.

Abstract

Folat, suda çözünebilen önemli bir B vitaminidir. Bitkisel ve hayvansal kaynaklı gıdalarda doğal olarak bulunmaktadır. Folat, metabolizmada önemli görevler üstlenmektedir. Bunlardan biri de plazma homosistein düzeyinin ayarlanmasıdır. Homosistein metabolizmasında görevli olan MTHFR (Metilentetrahidrofolat redüktaz) aktivitesinde azalma, homosistein seviyesini etkilemektedir. Bu nedenle MTHFR folat metabolizmasında önemli bir enzimdir. MTHFR geninde meydana gelen bazı mutasyonlar çeşitli hastalıklar için risk faktörü olan hiperhomosisteinemi ve homosisteinüriye sebep olabilmekte, metabolik sorunlara yol açabilmektedir. MTHFR, DNA sentezi, metilasyon reaksiyonları ve RNA sentezi gibi önemli yolaklarda etkilidir. MTHFR geninde meydana gelen en yaygın polimorfizmler C677T ve A1298C' dir. Bu polimorfizmlerin sıklığı populasyonlarda farklılık göstermektedir. MTHFR, folat dağılımını, homosistein metabolizmasını ve S-adenosilmetionin vasıtasıyla metilasyonu etkilediği için MTHFR' de genetik kusurlar hastalık riskini negatif ve pozitif olarak ayarlama potansiyeline sahiptir.
