The effect of the GM-CSF on fracture healing in rats

Abstract

Purpose: There are some relationship between osteogenesis and haematopoiesis. GM-CSF is a haematopoietic growth factor that is used in patients on myelosuppressive cancer therapy with neutropenia and aplastic anemia and skin ulcers. GM-CSF has stimulatory effects on especially granulocytes and macrophages lines. Study design: In this study the effect of GM-CSF on fracture healing in rats was investigated. Thirty rats with tibial diaphyseal fractures were divided into two groups. 1microgr/kg/day GM-CSF (Leucomax) was injected s.c to the first group for ten days while the second group had no treatment. Rats were sacrified at the end of the sixth week and evaluated radiologically and histopathologically. Modified Lane and Sandhu criteria were used to compare the results between study and control groups. Statistical analysis was performed with Mann-Whitney U test. Results: Radiologic evaluation showed that bone formation and union were superior in the study group: (p=0.0277 and p=0.0143, respectively). Histopathologic scores for GM-CSF were superior to those control group : (p=0.0384, p=0.0008 and p=0.040 respectively). In our opinion that GM-CSF may be helpful on bone healing but more comprehensive and large studies are needed.
