Sığırlarda Gebelikle İlişkili Glikoproteinlerin Tespitinde Kullanılan İki Test Kitinin Karşılaştırılması

Abstract

Bu çalışmada;
sığırlarda bir gebelik biyo-markırı olan gebelik ilişkili glikoproteinlerin kan
plazmasında tespitine yönelik hazırlanan farklı iki ticari gebelik tanı kitinin
(Sığır Gebelik Test Kiti ve Görsel Gebelik Test Kiti) erken gebelik tanısındaki
etkinliklerinin karşılaştırılması amaçlandı. Bu amaçla inek (n=32) ve
düvelerden (n=37) tohumlama sonrası 27-30. günde kan örneği alındı. Kan
örneklerinden toplanan plazma örnekleri, Sığır Gebelik Test Kiti ve Görsel
Gebelik Test Kitleriyle üretici firma talimatlarına göre analiz edildi. İnek ve
düvelerin gebelik durumları kan örneklerinin alımından 10 gün sonra transrektal
ultrasonografi ile tespit edildi. Gebeliklerin devamlılığı ise tohumlama
sonrası 60. günde yapılan ikinci transrektal ultrasonografi ile teyit edildi.
Sığır Gebelik Test Kiti ve Görsel Gebelik Test Kitlerinin sırasıyla;
sensitiviteleri %80 - %80, spesiviteleri %91.2 - %85.3, pozitif prediktif
değerleri %90.3 - %84.8, negatif prediktif değerleri %81.6 - %80.6, ve doğruluk
oranları %85.5 - %82.6 oldu. Sonuç olarak; Sığır Gebelik Test Kiti ve Görsel
Gebelik Test Kitinin inek ve düvelerde erken gebelik tanısında etkinliklerinin
benzer olduğu ve rutin erken gebelik tespitinde güvenle kullanılabileceği
kanısına varıldı.

Comparison of Two Test Kits for the Detection of Pregnancy Associated Glycoproteins in Cattle

Abstract

In this study, it was aimed to
compare the efficiency of two commercial pregnancy test kits (Bovine Pregnancy
Test Kit and Visual Pregnancy Test Kit) based on detection of pregnancy
associated glycoproteins, which are pregnancy bio-markers in cattle for early
pregnancy diagnosis in the blood plasma of cattle. For this purpose blood
samples from dairy cows (n=32) and heifers (n=37) were collected between 27 and
30 days after insemination. Plasma samples
collected
from blood samples were analyzed with Bovine Pregnancy Test Kit and Visual
Pregnancy Test Kit according to manufacturer’s protocols. Pregnancy status of
cows and heifers was diagnosed ten days after blood samples collection by using
transrectal ultrasonography. The pregnancy was confirmed by second transrectal
ultrasonography at 60 days post insemination. The sensitivity, specificity,
positive predictive value, negative predictive value and accuracy rates of the
Bovine Pregnancy Test Kit and Visual Pregnancy Test Kit were 80% - 80%, 91.2% -
85.3%, 90.3% - 84.8%, 81.6% - 80.6%, and 85.5% - 82.6%, respectively. As a
result; it was concluded that Bovine Pregnancy Test Kit and Visual Pregnancy Test
Kit had similar efficacies in early pregnancy diagnosis and could be used
safely for routine early pregnancy diagnosis in cows and heifers.
