Purification of bovine liver cytoplasmic aldehyde dehydrogenase (aldh.) enzyme, investigation of its kinetic properties and inhibition by disulfiram

Abstract

Aldehyde dehydrogenase (Aldehyde oxidoreductase E.C. 1.2.1.3) is an enzyme that catalyzes the cravings of many aldehyde structures together with acetaldehyde formed as a result of alcohol metabolism. This enzyme has an important function in alcohol metabolism and in the emergence of the alcoholic type. Therefore, this cytoplasmic aldehyde dehydrogenase enzyme was purified from bovine heart, its kinetic properties at changed pHs and the nature of its inhibition by disulfiram, which is widely used in the treatment of alcoholics, were investigated. As a result, it was determined that the enzyme showed a higher activity at high pHs and disulfiram was a severe inhibitory substance for the enzyme.

Abstract

Aldehit dehidrogenaz (Aldehit oksidoredüktaz E.C. 1.2.1.3), alkol metabolizması sonucu oluşan asetaldehit ile birlikte birçok aldehit yapısının isteklerini katalize eden bir enzimdir. Bu enzimin alkol metabolizmasında ve alkolik tipin ortaya çıkmasında önemli bir işlevi vardır. Bu nedenle, bu sitoplazmik aldehit dehidrogenaz enzimi sığır kalbinden saflaştırıldı, değişen pH'lardaki kinetik özellikleri ve alkoliklerin tedavisinde yaygın olarak kullanılan disülfiram tarafından inhibisyonunun doğası araştırıldı. Sonuç olarak, enzimin yüksek pH'larda daha yüksek bir aktivite gösterdiği ve disülfiramın enzim için ciddi bir inhibitör madde olduğu belirlendi.
