In this paper, pseudocholinesterase, activity which is being determined by various colorimetric methods in our laboratory, is investigated by a manometric method. For this Durvose Warburg Apparatus is used. First, the reaction flask-manometer systems used in the experiments are calibrated with mercury, and the flask constants are calculated. By using reagent blank and acetic acid standard the precision and the accuracy of the method are calculated. Frech sera obtained from 10 healthy persons are tested, and the enzyme activity is found between 51 - 212 p. mole/hour/ml. Precision of the method was 8,7 % (as the coefficient of variation) and accuracy 2,64 (as the mean value of the deviation from real value) .
Bu çalışmada laboratuarımızda çeşitli kolorimetrik metotlarla tayin edilmekte olan psödokolinesteraz aktivitesi, manometrik metotla denendi. Bunun icin Warburg cihazı kullanıldı. Önce deneyde kullanilan reaksiyon-şişesi-manometre sistemlerinin cıva ile kalibrasyonu yapılıp şişe sabiteleri hesanlandı. Ayıraç körü ve asetik asit standardı ile çalışılarak metodun isabet (precision) ve doğruluk derecesi (accuracy) tayin edildi. Normal 10 şahistan alınan taze serumlarla yapılan çalışmalarda enzim aktivitesi 51 - 212 U mol/saat/ml. araslnda bulundu. Literatürde benzeri metotlarla normal sinlrlar 90 - 140 ünite ve 140 - 200 ünite bulunmuştur.
