Pharmacological Aspects of the Adrenal Medulla

Abstract

The references have been reviewed concerning the synthesis, uptake, storage and liberation of catecholamines from the adrenal medulla and the influence of drugs on these phenomena. The synthesis of catecholamines in the adrenal medulla resembles that occurring at the sympathetic nerve endings. However, noradrenaline is partly converted to adrenaline in the medulla unlike the sympathetic nerve endings. It is possible that the two catecholamines are stored in the separate chromaffin cells. The origin of the catecholamines in the medulla is in part those taken up from the circulating blood. The uptake of the other phenylethylamine derivatives also occurs in the chromaffin cells of the adrenal medulla. The drugs with quite different pharmacological properties exert the common property of liberating catecholamines from the medulla. The drugs release catecholamines by a direct action or by a reflex action via the central nervous system. Adrenaline and noradrenaline may be released by the drugs in fairly a selective manner. The chromaffin cells have muscarinic receptors in addition to the clasically known nicotinic receptors.

Farmakoloji Yönünden Sürrenal Medüllası II. Kateşolaminlerin Açığa Çıkmasi (Liberasyon)

Abstract

Sürrenal medüllasında kateşolaminlerin sentezi, tutulması, depo edilmesi ve liberasyonu ile ilgili olaylar ve bunlar üzerine ilaçların tesirine ilişkin literatür gözden geçirildi. Sürrenal medüllasında kateşolaminlerin sentezi postganglioner sempatik sinir ucunda cereyan edene benzer tarzda olmaktadır. Ancak medüllada, sempatik sinir uçlarının aksine, noradrenalin kısmen adrenalin'e dönüşür. Adrenalin ve noradrenalin'in ayrı hücreler içinde bulunması muhtemeldir. Sürrenal medüllasındaki kateşolaminlerin menşei, kısmen, dolaşan kandan alınıp depo edilen kateşolaminlerdir. Sürrenal medüllası, kateşolaminlerden başka, diğer bazı feniletilamin türevlerini de tutabilir. Tesirleri itibariyle farklı farmakolojik gruplara giren birçok ilaçlar kateşolaminlerin medülla tarafından tutulmasını inhibe etmek özelliğine sahiptirler. İlâçlar direkt tesirle veya santral sinir sistemi aracılığı ile sürrenal medüllasından kateşolamin liberasyonu yapabilirler. Bazı ilâçlar veya tenbih şekilleri daha ziyade noradrenalin, diğer bazıları ise daha ziyade adrenalin liberasyonuna sebep olmaktadırlar. Sürrenal medüllasında sadece nikotinik reseptörler değil fakat ilâveten müskarinik tipte kolinerjik reseptörler de mevcuttur.
