The Effect of Tween-60 (Polyoxyethylene Sorbitan Monostearate) Inducing Hyperplasia on Mouse Skin

Abstract

Dipole tip promotor maddelerden olan Tween-60 ın tekrarlayıcı tatbiklerinin fare derisinde meydana getirdiği hiperplazi etkisi araştırıldı ve neticeler literatürdekilerle karşılaştırıldı.
Tween-60 ın sebep olduğu epidermal hücre değişiklikleri bilhassa diferansiye hücrelerde asırı bir çoğalmayı sağlıyordu. Bunu temin eden en az tatbikat sayısı 2-3 gün veya tatbikat olarak bulundu. 6 gün tatbikatla epidermiste (+ +++) şiddetinde bir hiperplazi teessüs etti. Bu hiperplazi diffüz, muntazam ve hücrelerde şişme ödem, hücreler arası mesafelerde genişleme, köprücükler teşekkülü ile karakterli idi. Tween-60 tatbikatının kesilmesi ile bu hiperplazinin 2--4 gün sonra tedricen gerilemeye başladığı ve 7-10 gün içinde kaybolarak epidermisin normal histolojik görünümünü aldığı tespit edildi. Neticede, başlangıçta günlük tatbikatlarla epidermiste 3--4 günde iyi bir hiperplazinin meydana getirilebileceği ve bunun 22 3 günlük fasılalarla yapılacak tek Tween-60 tatbikatları ile ayni derecede muhafaza edilebileceği kanısına varıldı. Bu neticenin deneysel deri karsinogenezi çalışmalarında Promotion olayını tetkik etmede metod yönünden önemli olabileceğj düşünüldü.

Tween-60 (Polyoxyethylene Sorbitan Monostearate) Fare Derisinde Hiperplazi Doğuran Etkisi

Abstract

Hyperplastic effect of repeated applications of Tween-60 onto the mouse skin has been investigated, and results compared with those reported in the literatüre.
The cellular changes in the epidermis were consisted of marked proliferation of the differentiated spinal cells. The least number of the application causing hiperplasia of epidermis was found to be 2-3 times. 6 applications were produced ( I I + ) severe epi„ dermal hiperplasia. This was diffuse, regular, and characterised by cellular swelling, edema, expansion in the intercellular spaces, and also exageration of the intercellular bridges. With the interruption of Tween-60 applications, this hyperplasia was regressed gradually and disappeared through seventh to tenth days, and the histologic appearence of the epidermis was returned to mal. In this way, it is said that, a nice epidermal hyperplasia can be produced by daily application of Tween-60 within 3-4 days. Its severity can be keep up at the same level for a long time by single application every other day or 3 days interval. This finding was found to be important in respect to method of determination of promotion in the studies on experimental carcinogenesis.
